Neuromuscular Blocking Agents

Neuromuscular blocking agents are pharmacological drugs that temporarily interrupt communication between motor nerves and skeletal muscles, producing controlled muscle relaxation. In pharmacology, nondepolarizing agents competitively inhibit nicotinic acetylcholine receptors at the neuromuscular junction, while depolarizing agents activate these receptors and maintain membrane depolarization, preventing effective action-potential transmission and muscle contraction. Clinicians use these drugs to facilitate endotracheal intubation, support controlled ventilation, and improve surgical conditions. Because they cause paralysis without providing unconsciousness or pain relief, their use requires appropriate anesthesia, respiratory support, and monitoring of neuromuscular function.

Measuring Neuromuscular Junction Functionality

A functional assessment of the neuromuscular junction (NMJ) can provide essential information on the communication between muscle and nerve. Here we describe a protocol to comprehensively evaluate both the NMJ and muscle functionality using two different muscle-nerve preparations, i.e. soleus-sciatic and diaphragm-phrenic.

Deep Neuromuscular Blockade Leads to a Larger Intraabdominal Volume During Laparoscopy

This article presents a method for measuring the working abdominal volume during laproscopic myomectomy using the surgical grasper, and applies this technique to obtain pilot data on whether deep neuromuscular blockade (NMB) can enable the use of lower insufflation pressure. Reduced insufflation pressure during laproscopic surgery has been shown to reduce post-operative pain, and thus the use of NMB during surgery may enable improved patient outcomes.

Preparation and In Vitro Characterization of Dendrimer-based Contrast Agents for Magnetic Resonance Imaging

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.
