Hardy-Weinberg and Genetic Drift

Per capire questa equazione, torniamo all'esempio della giraffa. La A maiuscola rappresenta l'allele tan poiché è dominante, e la a minuscola è per l'allele bruno perché è recessivo. La frequenza di questi due alleli nella popolazione è designata rispettivamente come p e q. Quindi, come facciamo a conoscere la frequenza allelica? Bene, ogni individuo ha due alleli. In questo esempio, il 40% degli alleli nel pool genetico sono abbronzati. Pertanto, la frequenza dell'allele tan, p, è 0,4 e la frequenza dell'allele marrone, q, è 0,6. Si noti che p più q è sempre uguale a uno. 
Ora torniamo all'equazione di Hardy-Weinberg. Ogni termine nell'equazione rappresenta una frequenza del genotipo. La frequenza del genotipo omozigote dominante è p al quadrato e l'omozigote recessivo è rappresentato da q al quadrato. Il genotipo eterozigote è due pq. La ragione per cui moltiplichiamo per due qui è che ci sono due modi diversi di generare un genotipo eterozigote. Combinati, questi rappresentano tutti il 100% dei genotipi. Quindi, una frequenza totale di uno. Per mantenere questo equilibrio, il principio dell'equilibrio di Hardy-Weinberg afferma che una popolazione dovrebbe soddisfare cinque ipotesi principali. Ci dovrebbero essere accoppiamenti casuali, grandi dimensioni della popolazione, nessuna mutazione, nessuna selezione sul gene in questione e nessun flusso genico dentro o fuori la popolazione. Un malinteso generale sull'evoluzione è che richieda la selezione naturale per avvenire. Tuttavia, non è sempre così. La deriva genetica è un meccanismo attraverso il quale l'evoluzione può avvenire senza la selezione naturale. È definito come un cambiamento nella frequenza allelica di una popolazione dovuto al caso. Per immaginare questo, torniamo all'esempio di una popolazione di giraffe e immaginiamo che i loro alleli di marrone chiaro e marrone siano rappresentati da biglie di due colori diversi. Supponiamo qui che ogni colore inizi ugualmente abbondante. Se dovessimo iniziare una nuova generazione di questa popolazione, avremmo bisogno di allevare coppie di individui e quindi selezionare tra quattro alleli per coppia. Se selezioniamo una coppia riproduttiva a caso, potremmo ritrovarci con due biglie di ogni colore. Tuttavia, solo per caso, alcuni abbinamenti avranno un solo colore marmo, oppure tre di un colore e uno dell'altro. Queste deviazioni casuali da 50-50 su più accoppiamenti per creare una nuova generazione potrebbero significare che la generazione successiva non ha più una miscela uguale di ciascun allele.

Pertanto, a differenza dell'evoluzione adattativa, in cui la frequenza allelica cambia per selezionare i tratti adatti all'ambiente, come le coccinelle con una maggiore quantità di melanina che sopravvivono meglio nei climi più freddi a causa di una migliore capacità di assorbire il calore, la deriva genetica rappresenta un tipo di evoluzione che è puramente dovuta al cambiamento stocastico. Ad esempio, la rimozione casuale di una parte di una popolazione attraverso un evento catastrofico.

Il cambiamento evolutivo è interessante e importante da studiare, ma i cambiamenti nelle popolazioni si verificano per lunghi periodi di tempo e in enormi spazi fisici e sono quindi molto difficili da misurare. In generale, lo studio di fenomeni di questo tipo richiede l'uso di modelli matematici costruiti utilizzando parametri che possono essere convenientemente misurati. Questi modelli vengono quindi utilizzati per fare previsioni su come le modifiche al sistema potrebbero influenzare il risultato. 
Ad esempio, i cambiamenti nella frequenza degli alleli genetici nei singoli loci nelle specie sono spesso osservati per lunghi periodi di tempo, ma non sono generalmente osservabili per brevi periodi di tempo. L'uso di modelli computerizzati consente ai ricercatori di prevedere i cambiamenti all'interno del pool genetico di una specie utilizzando osservazioni già raccolte e consente simulazioni di un numero potenzialmente illimitato di generazioni della popolazione. Questo non sarebbe certamente possibile nell'arco di una vita umana.
<h4>Il principio di equilibrio di Hardy-Weinberg</h4>
Un'equazione utilizzata per modellare le popolazioni è l'equazione di equilibrio di Hardy-Weinberg. È stato formulato indipendentemente nel 1908 sia da G. H. Hardy che da Wilhelm Weinberg1,2. La semplice equazione descrive la frequenza allelica attesa di una popolazione che non è in evoluzione. Ogni espressione nell'equazione rappresenta la frequenza prevista di uno dei tre possibili genotipi diploidi. La frequenza dominante omozigote è rappresentata da p2, omozigote recessiva da q2 ed eterozigote da 2pq. Se queste frequenze vengono sommate, la somma è 1, il che ha senso dal momento che l'intera popolazione è stata divisa nelle tre categorie disponibili.
È anche possibile descrivere la popolazione, o pool genico, in termini dei due alleli, indipendentemente da come sono impacchettati in individui diploidi. Questa equazione è rappresentata come p + q = 1, cioè le frequenze degli alleli dominanti e recessivi devono sommarsi fino a 1 all'interno della popolazione. Ancora una volta, questo ha senso, dal momento che il modello include solo due possibilità alleliche. Si noti che la frequenza dei genotipi nella popolazione è solo l'espansione quadratica della frequenza degli alleli nella popolazione perché (p + q)2 = p2 + 2pq + q2.
L'equazione di Hardy-Weinberg, come la maggior parte degli altri modelli, richiede una serie di ipotesi. Utilmente, se i dati della vita reale differiscono dai dati previsti, è possibile ipotizzare quale delle ipotesi di partenza fosse falsa. Le ipotesi delle equazioni di equilibrio di Hardy-Weinberg sono: 1) la popolazione è molto grande, 2) la popolazione è chiusa, il che significa che non ci sono individui che immigrano o emigrano fuori dalla popolazione, 3) non ci sono mutazioni che si verificano sul gene in questione, 4) gli individui all'interno della popolazione si accoppiano a caso: gli individui non scelgono i loro compagni, 5) La selezione naturale non è in corso. Ancora una volta, tieni presente che l'equazione di equilibrio di Hardy-Weinberg è un'ipotesi nulla. Spesso, le popolazioni reali violano una o più di queste ipotesi. L'equazione viene utilizzata per determinare se e come sta avvenendo l'evoluzione.
<h4>Deriva genetica</h4>
In una popolazione relativamente piccola, una condizione che viola la prima ipotesi di Hardy-Weinberg, è possibile che le frequenze alleliche siano il risultato del caso. Questo fenomeno è indicato come deriva genetica. Una versione di questo è indicata come effetto fondatore. Se un piccolo numero di individui si sposta in un luogo isolato e inizia una nuova popolazione, la genetica specifica di quei particolari individui plasmerà le generazioni future. Questo nuovo piccolo pool genetico può avere la stessa frequenza allelica dell'originale, ma è anche possibile, persino probabile, che non lo faccia. Diciamo che la popolazione originale comprendeva il 50% di alleli dominanti e il 50% di recessivi. All'estremo, se tutti gli individui in un gruppo fondatore migrante sono omozigoti recessivi, allora l'allele dominante è stato perso completamente e il recessivo è ora al 100%. Un fenomeno simile, chiamato effetto collo di bottiglia, può verificarsi se una popolazione subisce un forte calo numerico a causa di un disastro naturale, di un intervento umano o di una malattia.
È importante sottolineare che la deriva genetica rappresenta un tipo di evoluzione che non è necessariamente adattiva. Non seleziona specificamente i tratti adatti all'ambiente. Tuttavia, è un'importante forza evolutiva nel plasmare popolazioni con un piccolo numero di individui e causerà deviazioni dalle previsioni di Hardy-Weinberg. Nelle popolazioni più grandi, è più probabile che le deviazioni dalle previsioni significhino che la popolazione in questione sta subendo un'evoluzione attraverso la selezione naturale. In queste circostanze, si può esplorare un'ulteriore considerazione dei meccanismi che causano questa evoluzione.
Le moderne applicazioni del modello di Hardy-Weinberg includono l'analisi delle popolazioni umane e animali in termini di come i loro sistemi immunitari si relazionano alla loro suscettibilità alle malattie infettive3,4. Molti gruppi di ricerca stanno catalogando geni che codificano specifiche molecole del sistema immunitario, come CCR5 e il complesso maggiore di istocompatibilità (MHC, noto nell'uomo come antigene leucocitario umano (HLA), e correlando queste informazioni a studi epidemiologici riguardanti la malattia e la sua progressione3,5-6. Dal confronto di questi due tipi di set di dati, stanno emergendo modelli che mostrano che alcuni individui sono geneticamente resistenti a particolari infezioni, mentre altri hanno maggiori probabilità di soccombere alla malattia4. Hardy-Weinberg tests help scientists check whether a population is changing over time. Because evolution happens slowly and over large areas, researchers often use mathematical models to study it. These models use measurable data and computer simulations to predict how allele frequencies may shift across many generations.

The Hardy-Weinberg equilibrium equation is one common model. It was developed independently in 1908 by G. H. Hardy and Wilhelm Weinberg. The equation gives the expected allele frequency for a population that is not evolving, so it serves as a useful null hypothesis. If measured allele frequencies do not match the prediction, the gene may be undergoing evolutionary change.

The model is written as p2 + 2pq + q2 = 1. In this equation, p is the frequency of the dominant allele and q is the frequency of the recessive allele. The three terms represent the expected frequencies of the three diploid genotypes: homozygous dominant, heterozygous, and homozygous recessive. The allele version of the model is p + q = 1, since the two allele frequencies must total 1 in the population.

Hardy-Weinberg also depends on a set of assumptions. The population must be very large and closed, which means no individuals move in or out. There must be no mutations in the gene being studied. Individuals must mate at random, and natural selection must not be acting. When real data breaks one or more of these assumptions, the model can help point to what may be happening.

Genetic drift is one reason a small population can change by chance alone. This violates the large-population assumption of Hardy-Weinberg. One form of genetic drift is the founder effect. It happens when a small group leaves and starts a new isolated population. The genes carried by those founders shape future generations. In an extreme case, if the founders are all homozygous recessive, the dominant allele can disappear and the recessive allele can reach 100%.

A similar pattern is called the bottleneck effect. It can occur after a natural disaster, human intervention, or disease causes a major drop in population size. Genetic drift is not necessarily adaptive, because it does not specifically favor traits that fit the environment. Even so, it is an important evolutionary force in small populations and can create differences from Hardy-Weinberg predictions. In larger populations, those differences are more likely to suggest natural selection.

Scientists also use Hardy-Weinberg testing in modern studies of human and animal populations. One focus is how immune system genes relate to susceptibility to infectious disease. Researchers study genes such as CCR5 and the major histocompatibility complex, or MHC, called HLA in humans. By comparing genetic data with disease data, they can find patterns showing that some individuals are more resistant to certain infections while others are more likely to become ill. These studies have helped explain how infections have influenced evolution.

Content elaboration: Natural selection is used to describe the process by which traits become more or less common in a population due to consistent environmental pressures upon the survival and reproduction of individuals with the trait. Mathematical reasoning is applied to solve problems (e.g., use Hardy-Weinberg principle to explain deviations in observed gene frequency patterns in a population compared to expected patterns based on the assumptions of the principle). Populations evolve over time. Evolution through natural selection is the consequence of the interactions of:
1. The potential for a population to increase its numbers; 
2. The genetic variability of offspring due to mutation and recombination of genes; 
3. A finite supply of the resources required for life; and 
4. The differential survival and reproduction of individuals based on phenotype(s). 
Mutations are described in the content elaboration for Heredity. Apply the knowledge of mutation and genetic drift to real-world examples. Biological evolution explains the natural origins for the diversity of life. Emphasis shifts from thinking in terms of selection of individuals with a particular trait to changing proportions of a trait in populations as a result of the mechanisms of natural selection, genetic drift, movement of genes into and out of populations and sexual selection.

Classification systems are frameworks, developed by scientists, for describing the diversity of organisms; indicating the degree of relatedness among organisms. Recent molecular sequence data generally support earlier hypotheses regarding lineages of organisms based upon morphological comparisons. Both morphological and molecular comparisons can be used to describe patterns of biodiversity (cladograms present hypotheses to explain descent from a common ancestor with modification). The concept of descent from a common ancestor with modification provides a natural explanation for the diversity of life on Earth as partially represented in the fossil record and in the similarities of existing species. 
Variation of organisms within a species due to population genetics and gene frequency
Different phenotypes result from new combinations of existing genes or from mutations of genes in reproductive cells. At the high school level, the expectation is to combine grade 8 knowledge with an explanation of genes and the function of chromosomes. Natural selection works on the phenotype. Heritable characteristics influence how likely an organism is to survive and reproduce in a particular environment. When an environment changes, the survival value of inherited characteristics may change. This may or may not cause a change in species that inhabit the environment. Use real-world examples to illustrate natural selection, gene flow, sexual selection, and genetic drift.
