Ti sei mai chiesto perché ogni autunno le foglie degli alberi cambiano dal verde alle sfumature del giallo, dell'arancione e del rosso? Cosa dà alle foglie i loro colori vivaci in primo luogo? La risposta sta negli organelli vegetali chiamati cloroplasti, che contengono pigmenti che assorbono determinate lunghezze d'onda alla luce del sole e ne riflettono altre. Un pigmento particolare, la clorofilla, è il più abbondante in estate. Assorbe le lunghezze d'onda ad alta energia, viola, blu e rosse dalla luce solare e riflette le lunghezze d'onda verdi conferendo alle foglie il loro aspetto verde. Ci sono altri pigmenti nelle foglie, come i carotenoidi, che riflettono la luce rossa e gialla. In autunno le foglie smettono di reintegrare i loro pigmenti. Poiché la clorofilla si degrada più velocemente degli altri pigmenti, i colori di questi carotenoidi vengono smascherati.La presenza di diversi pigmenti in una foglia verde può essere dimostrata con la carta per cromatografia, un polimero idrofilo che separa le molecole in base alla loro solubilità in un particolare solvente. Il valore Rf è il rapporto tra la distanza percorsa da un pigmento e la distanza percorsa dal solvente. Ogni pigmento ha un valore Rf univoco e possiamo facilmente identificare i pigmenti confrontando i valori calcolati con gli standard. La fotosintesi, il processo mediante il quale le piante convertono l'anidride carbonica, l'acqua e l'energia luminosa in energia chimica e ossigeno, viene effettuata principalmente nelle foglie di una pianta e la clorofilla svolge un ruolo fondamentale in questo processo. Ad alti tassi di fotosintesi, l'acqua viene scissa più velocemente per reintegrare gli elettroni e l'ossigeno viene generato rapidamente.Questo fenomeno ci aiuta a valutare il tasso di fotosintesi in laboratorio semplicemente sospendendo i dischi fogliari in una soluzione di bicarbonato in cui il bicarbonato agisce come una ricca fonte di carbonio. All'inizio dell'esperimento del disco fogliare, i gas vengono espulsi dai dischi fogliari applicando una pressione negativa con un vuoto in una siringa. I dischi fogliari con i loro gas espulsi diventano più pesanti e affondano sul fondo della soluzione di bicarbonato quando vengono trasferiti in un becher. Quando avviene la fotosintesi, l'acqua nell'ambiente viene scissa per reintegrare gli elettroni della clorofilla. L'ossigeno risultante rende i dischi più leggeri facendoli galleggiare in superficie nel tempo. Gli ambienti che consentono tassi più elevati di fotosintesi hanno dischi che galleggiano più velocemente.In questo laboratorio separerai e identificherai prima i pigmenti nelle foglie di spinaci usando la carta per cromatografia. Quindi valuterai il tasso di fotosintesi in acqua e in soluzione di bicarbonato con un esperimento sul disco fogliare.

<h4>Autotrofi</h4>

Quasi tutti gli organismi viventi sulla Terra dipendono dalla fotosintesi, che è il processo che converte l'energia della luce solare in uno zucchero semplice chiamato glucosio. Gli autotrofi sono organismi che catturano l'energia luminosa utilizzando la fotosintesi. Conosciuti anche come produttori primari, forniscono l'energia necessaria agli organismi che li mangiano, noti come consumatori.

Gli organismi in grado di fotosintetizzare sono piuttosto diversi, tra cui i cianobatteri, alcuni protisti come le alghe e le piante. Nelle cellule eucariotiche, la fotosintesi avviene in un organello chiamato cloroplasto, che appare verde a causa del suo alto contenuto di pigmento clorofilla. I pigmenti sono molecole che assorbono la luce a determinate lunghezze d'onda. La luce che non viene assorbita da un pigmento viene riflessa come luce visibile che può essere osservata come il colore del pigmento. Le piante producono più pigmenti con varie funzioni che assorbono la luce dal sole a diverse lunghezze d'onda. Ad esempio, la clorofilla assorbe la luce alle lunghezze d'onda del rosso e del blu mentre riflette le lunghezze d'onda che corrispondono al verde. Alcuni altri pigmenti come carotenoidi, antociani e betalaine riflettono comunemente la luce a lunghezze d'onda a bassa energia tra 600 e 800 nm e quindi appaiono dal giallo al rosso. Nelle regioni temperate, la diminuzione della clorofilla in autunno rivela questi pigmenti quando le foglie cambiano colore in rosso, giallo e arancione.

<h4>Cloroplasti</h4>

La clorofilla è il pigmento principale nei cloroplasti utilizzati nella fotosintesi, mentre altri pigmenti aiutano a incanalare l'energia luminosa verso le clorofille o a proteggere la cellula dai danni della luce. La fotosintesi consiste di due percorsi, noti come reazioni dipendenti dalla luce e indipendenti dalla luce e si verificano in posizioni distinte all'interno dei cloroplasti. Questi organelli contengono tre membrane: una membrana esterna, una membrana interna e la membrana tilacoide più interna, che forma lunghe pieghe a forma di disco all'interno del cloroplasto. Lo spazio pieno di liquido tra le membrane interne e tilacoidi è chiamato stroma. Le reazioni dipendenti dalla luce iniziano quando l'energia della luce solare eccita gli elettroni dei pigmenti clorofilliani che sono incorporati all'interno della membrana tilacoidale. Questi elettroni ad alta energia vengono quindi passati da una molecola di trasporto di elettroni all'altra all'interno della membrana tilacoidale, nota collettivamente come catena di trasporto degli elettroni. Ogni trasferimento all'interno della catena di trasporto dell'elettrone porta l'elettrone a uno stato energetico inferiore, rilasciando quindi energia. Parte di questa energia viene sfruttata per sintetizzare piccole molecole ricche di energia come l'adenosina trifosfato (ATP) e la nicotinammide adenina dinucleotide fosfato (NADPH). L'elettrone perso della clorofilla viene sostituito attraverso l'idrolisi (che significa scissione dell'acqua), quando gli atomi di idrogeno e ossigeno vengono separati. Gli atomi di idrogeno generati dall'idrolisi donano i loro elettroni alla clorofilla mentre le molecole di ossigeno vengono rilasciate nell'atmosfera. Il flusso di elettroni attraverso l'idrolisi consente ai pigmenti della clorofilla di assorbire continuamente l'energia luminosa eccitando ripetutamente nuovi elettroni e passandoli lungo la catena di trasporto degli elettroni.

Le reazioni indipendenti dalla luce avvengono nello stroma. Questo processo utilizza l'energia luminosa indirettamente utilizzando l'energia delle molecole di ATP e NADPH generata dalle reazioni dipendenti dalla luce. Durante questo percorso, l'anidride carbonica (CO2) viene utilizzata per costruire uno zucchero a tre atomi di carbonio, che può poi essere trasformato in glucosio o altre biomolecole. Questo processo è noto anche come fissazione del carbonio o sequestro del carbonio, perché il carbonio viene sequestrato dall'atmosfera e fissato in biomolecole.

<h4>La fotosintesi e l'ambiente</h4>

Il sequestro del carbonio durante la fotosintesi è una fase importante del ciclo del carbonio, in cui la CO2 fluisce da un serbatoio all'altro a un ritmo relativamente costante. Le variazioni della portata di CO2 possono spostare l'equilibrio tra i serbatoi. Ancora più importante, poiché la CO2 è un gas serra, l'aumento della sua concentrazione atmosferica contribuisce all'aumento delle temperature. La maggior parte di questa CO2 viene emessa dalla combustione di combustibili fossili, restituendo così l'anidride carbonica che è stata sequestrata dalla fotosintesi centinaia di milioni di anni fa nell'atmosfera a un ritmo senza precedenti. Pertanto, le foreste e le alghe oceaniche sono sempre più indispensabili per raffreddare la terra riducendo l'aumento dei livelli di CO2 1. Questo è uno dei tanti motivi per cui la deforestazione è una seria preoccupazione in un clima che cambia.

Proprio come le piante usano la fotosintesi per catturare l'energia luminosa in biomolecole, i ricercatori stanno studiando la fotosintesi artificiale per creare biocarburanti a zero emissioni di carbonio come alternativa ai combustibili fossili. Analogamente ai pannelli solari, i metodi di fotosintesi artificiale ottengono energia dal sole e la immagazzinano come energia chimica, parte della quale può essere immagazzinata per lunghi periodi di tempo, invece di convertirla direttamente in elettricità2. Gli scienziati sono stati anche in grado di produrre zuccheri semplici e acido lattico utilizzando batteri fotosintetici3. Questo approccio ha ampie applicazioni nella produzione di biomolecole, biocarburanti e persino plastiche biodegradabili con emissioni nocive minime o nulle. Inoltre, questo approccio può essere utilizzato per rimuovere l'eccesso di CO2 dall'atmosfera. Un concetto simile viene applicato in campo medico, dove i batteri fotosintetici vengono utilizzati per produrre sostanze e farmaci fisiologicamente attivi, alcuni dei quali sono stati applicati alla diagnosi e al trattamento del cancro4. La ricerca futura potrebbe aumentare l'efficienza della produzione di molecole sostenibili e a zero emissioni di carbonio, rimediando al contempo agli effetti dell'aumento dei livelli di CO2 nell'atmosfera.

photosynthesis: light, sugars, and carbon

Photosynthesis turns sunlight into chemical energy in the form of glucose. This process powers most life on Earth by helping autotrophs, or primary producers, make food. The glucose they make can provide quick energy or be stored as starch for later use. Consumers depend on these producers for energy.

Photosynthesis happens in many kinds of organisms. Cyanobacteria, some protists such as algae, and plants can all carry out this process. In eukaryotic cells, it takes place in the chloroplast. Chloroplasts look green because they contain chlorophyll, a pigment that absorbs light at specific wavelengths.

Plants use several pigments to capture light. Chlorophyll absorbs red and blue light and reflects green light. Other pigments, including carotenoids, anthocyanins, and betalains, absorb or reflect different wavelengths and often appear yellow to red. In autumn, when chlorophyll levels drop in temperate regions, these pigments become visible and leaves change color.

Photosynthesis has two main stages: light-dependent reactions and light-independent reactions. These stages happen in different parts of the chloroplast. The chloroplast has an outer membrane, an inner membrane, and an inner thylakoid membrane that forms disc-like folds. The space between the inner membrane and thylakoids is the stroma.

The light-dependent reactions begin in the thylakoid membrane when sunlight excites electrons in chlorophyll. Those electrons move through an electron transport chain, dropping to lower energy levels and releasing energy. The cell uses that energy to make ATP and NADPH. Water splitting, also called hydrolysis, replaces the lost electrons in chlorophyll. This process releases oxygen into the atmosphere and helps keep the flow of electrons going.

The light-independent reactions take place in the stroma. They use ATP and NADPH from the first stage to build a three-carbon sugar from carbon dioxide. That sugar can then be turned into glucose or other biomolecules. This is also called carbon fixation or carbon sequestration because carbon is pulled from the atmosphere and stored in living molecules.

Photosynthesis plays a major role in the carbon cycle. Carbon dioxide moves between reservoirs at a steady pace, but changes in that flow can upset the balance. Because carbon dioxide is a greenhouse gas, more of it in the atmosphere can raise temperatures. Much of the extra carbon dioxide comes from burning fossil fuels, which returns ancient carbon to the air very quickly.

Forests and ocean algae help lower carbon dioxide levels, so deforestation is a serious concern in a changing climate. Scientists are also studying artificial photosynthesis as a way to make carbon-neutral biofuels. Like solar panels, these methods take energy from the sun and store it as chemical energy instead of electricity. Researchers have also used photosynthetic bacteria to make simple sugars, lactic acid, biodegradable plastics, and even medically useful compounds, including substances studied for cancer diagnosis and treatment.
