Cellular Respiration

Tutto ciò che è vivo ha bisogno di una fonte di energia per alimentare le sue attività. In definitiva, quella fonte di energia è il sole. In che modo allora gli organismi sulla Terra hanno sfruttato questa energia? Tutto inizia con i fotosintetizzatori. Questi organismi sono in grado di prendere anidride carbonica e acqua e quindi utilizzare l'energia catturata dal sole sotto forma di fotoni per forzare queste molecole insieme, producendo glucosio e ossigeno. Il glucosio è la chiave per il prossimo passo critico, quello che quasi tutti gli organismi usano in una forma o nell'altra: la respirazione cellulare. La respirazione cellulare è effettuata da enzimi, sia nella membrana cellulare nei procarioti che nei mitocondri negli eucarioti.

La reazione chimica inizia scomponendo il glucosio utilizzando l'ossigeno per creare nuovamente anidride carbonica e acqua, ma nel processo, l'energia che originariamente è stata utilizzata per produrre la molecola di glucosio va in due nuovi posti. Uno è la sintesi di molecole di ATP, o adenosina trifosfato, una fonte di energia che le cellule possono facilmente utilizzare. Il resto si perde sotto forma di calore. Finora abbiamo parlato di un tipo di respirazione cellulare che utilizza l'ossigeno, e questa è indicata come respirazione aerobica, ma alcuni organismi e cellule sono in grado di fare respirazione in assenza di ossigeno. Questa è chiamata respirazione anaerobica. E invece di produrre CO2 e acqua, questo processo produce etanolo come sottoprodotto. La fermentazione è un esempio di questo tipo di respirazione. È così che il lievito è in grado di produrre alcol in contenitori sigillati. Possiamo usare uno strumento chiamato respirometro per misurare la respirazione aerobica. Molto semplicemente, questo dispositivo misura la quantità di ossigeno utilizzata da un organismo, in questo caso facendo germogliare i semi delle piante. I semi in germinazione respirano, il che significa che possiamo aspettarci che consumino ossigeno e rilascino anidride carbonica. Non hanno ancora le loro parti verdi, quindi non stanno fotosintetizzando.

La misurazione della respirazione in un respirometro utilizza un metodo intelligente basato sulla legge dei gas ideali, P per V è uguale a n volte R per T. P è la pressione del sistema. V è il volume del gas. n è il numero di moli di gas presenti. R è la costante di gas ideale... e T è la temperatura assoluta. In parole povere, ciò significa che è possibile capire quante molecole di gas sono presenti in un campione misurandone il volume, supponendo di conoscere la pressione e la temperatura, perché R è una costante.

I respirometri contengono idrossido di potassio, che intrappola l'anidride carbonica in forma solida come carbonato di potassio. Quindi, a causa della respirazione cellulare, l'ossigeno nel tubo sigillato viene utilizzato dai semi e rilasciano anidride carbonica, che a sua volta viene intrappolata come carbonato di potassio. Pertanto, con il progredire della respirazione cellulare, il volume totale di gas all'interno del sistema diminuisce. Possiamo quantificare questo collegando il respirometro a un dispositivo chiamato manometro. Qui, quando le molecole di ossigeno vengono consumate, la pressione scende all'interno della camera del respirometro e un liquido colorato all'interno di un minuscolo tubo capillare viene tirato nella direzione della pressione diminuita. Possiamo quindi stimare la quantità di gas rimanente nel tubo del respirometro leggendo il valore a livello del manometro. Questo versatile sistema può essere configurato con molte variabili diverse, come ad esempio temperature diverse, e può essere utilizzato per testare la frequenza respiratoria in molti tipi diversi di organismi viventi.

<h4>Autotrofi ed eterotrofi</h4>

Gli organismi viventi richiedono un apporto continuo di energia per mantenere le funzioni cellulari e dell'organismo come la crescita, la riparazione, il movimento, la difesa e la riproduzione. Le cellule possono utilizzare solo l'energia chimica per alimentare le loro funzioni, quindi hanno bisogno di raccogliere energia dai legami chimici delle biomolecole, come zuccheri e lipidi. Gli organismi autotrofi, vale a dire le piante, le alghe e i batteri fotosintetici e chemiosintetici, convertono i materiali inorganici in tali biomolecole sfruttando l'energia dall'ambiente, ad esempio dalla luce solare durante la fotosintesi. Gli organismi eterotrofi non sono in grado di sintetizzare biomolecole ad alta energia da materiali inorganici, quindi ottengono energia consumando composti di carbonio prodotti da altri organismi, principalmente da autotrofi. Quando è necessaria energia, i legami chimici dei composti del carbonio vengono rotti per raccogliere l'energia immagazzinata in questi legami. I processi per raccogliere energia dalle biomolecole sono chiamati respirazione cellulare.

La respirazione cellulare si verifica sia negli organismi autotrofi che in quelli eterotrofi, dove l'energia diventa disponibile per l'organismo più comunemente attraverso la conversione dell'adenosina difosfato (ADP) in adenosina trifosfato (ATP). Esistono due tipi principali di respirazione cellulare: la respirazione aerobica e la respirazione anaerobica. La respirazione aerobica è un tipo specifico di respirazione cellulare, in cui l'ossigeno (O2) è necessario per creare ATP. In questo caso, il glucosio (C6H12O6) può essere ossidato completamente in una serie di reazioni enzimatiche per produrre anidride carbonica (CO2 ) e acqua (H2O).

<h4>Respirazione aerobica</h4>

La respirazione aerobica avviene in tre fasi. Un processo chiamato glicolisi divide il glucosio in due molecole a tre atomi di carbonio chiamate piruvato. Questo processo rilascia energia, parte della quale viene trasferita all'ATP. Successivamente, le molecole di piruvato entrano nei mitocondri per prendere parte a una serie di reazioni chiamate ciclo di Krebs, noto anche come ciclo dell'acido citrico. Questo completa la scomposizione del glucosio, raccogliendo parte dell'energia in ATP e trasferendo gli elettroni sulle molecole trasportatrici. Nell'ultimo stadio, noto come fosforilazione ossidativa, gli elettroni passano attraverso un sistema di trasporto di elettroni nella membrana interna mitocondriale, che mantiene un gradiente di ioni idrogeno. Le cellule sfruttano l'energia di questo gradiente protonico per generare la maggior parte dell'ATP durante la respirazione aerobica.

<h4>Respirazione anaerobica</h4>

La respirazione aerobica richiede ossigeno, tuttavia, ci sono molti organismi che vivono in luoghi in cui l'ossigeno non è prontamente disponibile o dove altre sostanze chimiche travolgono l'ambiente. Gli estremofili sono batteri che possono vivere in luoghi come le bocche idrotermali oceaniche profonde o le grotte sottomarine. Piuttosto che utilizzare l'ossigeno per sottoporsi alla respirazione cellulare, questi organismi utilizzano accettori inorganici come il nitrato o lo zolfo, che sono più facilmente ottenibili in questi ambienti difficili. Questo processo è chiamato respirazione anaerobica.

Quando l'ossigeno non è presente e la respirazione cellulare non può avvenire, si verifica una speciale respirazione anaerobica chiamata fermentazione. La fermentazione inizia con la glicolisi per catturare parte dell'energia immagazzinata nel glucosio in ATP. Tuttavia, poiché non si verifica fosforilazione ossidativa, la fermentazione produce meno molecole di ATP rispetto alla respirazione aerobica. Nell'uomo, la fermentazione avviene nei globuli rossi che mancano di mitocondri, così come nei muscoli durante un'attività faticosa che genera acido lattico come sottoprodotto, quindi è chiamata fermentazione dell'acido lattico. Alcuni batteri svolgono la fermentazione dell'acido lattico e vengono utilizzati per realizzare prodotti come lo yogurt. Nel lievito, un processo noto come fermentazione alcolica genera etanolo e anidride carbonica come sottoprodotti ed è stato utilizzato dall'uomo per fermentare bevande o far lievitare l'impasto.

<h4>Fotosintesi</h4>

La respirazione cellulare insieme alla fotosintesi è una caratteristica del trasferimento di energia e materia e mette in evidenza l'interazione degli organismi con il loro ambiente e con gli altri organismi della comunità. La respirazione cellulare avviene all'interno delle singole cellule, tuttavia, su scala degli ecosistemi, lo scambio di ossigeno e anidride carbonica attraverso la fotosintesi e la respirazione cellulare influisce sui livelli di ossigeno atmosferico e anidride carbonica.

È interessante notare che i processi di respirazione cellulare e fotosintesi sono direttamente opposti l'uno all'altro, dove i prodotti di una reazione sono i reagenti dell'altra. La fotosintesi produce il glucosio che viene utilizzato nella respirazione cellulare per produrre ATP. Questo glucosio viene poi riconvertito in CO2 durante la respirazione, che è un reagente utilizzato nella fotosintesi. <h4>Misurazione della respirazione</h4>

Gli scienziati possono misurare il tasso di respirazione cellulare utilizzando un respirometro valutando il tasso di scambio di ossigeno. Comprendere la Legge dei Gas Ideali è di fondamentale importanza per sapere come funziona il respirometro. La legge dei gas ideali afferma che il numero di molecole di gas in un contenitore può essere determinato dalla pressione, dal volume e dalla temperatura. Più specificamente, il prodotto del volume e della pressione di un gas è uguale al prodotto del numero di molecole di gas, della costante ideale del gas e della temperatura del gas. I respirometri contengono idrossido di potassio che intrappola l'anidride carbonica prodotta dalla respirazione in forma solida come carbonato di potassio. In particolare, i mitocondri sono essenziali per la respirazione cellulare e qualsiasi condizione che influisce sulla salute dei mitocondri ha immense conseguenze per la salute dell'organismo. Ad esempio, le miopatie mitocondriali sono un gruppo di malattie neuromuscolari causate da danni mitocondriali, che colpiscono prevalentemente le cellule nervose e muscolari, che richiedono alti livelli di energia per funzionare1. Inoltre, molti veleni agiscono inibendo la respirazione cellulare. Ad esempio, il cianuro inibisce la produzione di ATP attraverso la fosforilazione ossidativa, quindi la comprensione dei meccanismi del cianuro o di altri veleni metabolici consente il trattamento di individui che sono stati esposti ad essi2. Allo stesso modo, alcuni farmaci come alcuni antibiotici, chemioterapici, statine e anestetici possono anche interferire con la funzione mitocondriale e potrebbero non essere adatti per il trattamento di pazienti che hanno disturbi mitocondriali3.

Cellular respiration is the process cells use to release usable energy from biomolecules such as sugars and lipids. Living things need a steady energy supply for growth, repair, movement, defense, and reproduction. Cells use that energy mainly by converting adenosine diphosphate (ADP) to adenosine triphosphate (ATP).

Autotrophs and heterotrophs get the molecules for respiration in different ways. Autotrophs, such as plants, algae, and photosynthetic and chemosynthetic bacteria, build high-energy biomolecules from inorganic materials by using energy from the environment, such as sunlight. Heterotrophs cannot make those molecules from inorganic materials, so they obtain carbon compounds by eating other organisms, mainly autotrophs.

Aerobic respiration needs oxygen to make ATP. In this form of respiration, glucose can be fully broken down through a series of enzyme-driven reactions into carbon dioxide and water. Aerobic respiration has three stages: glycolysis, the Krebs cycle, and oxidative phosphorylation.

Glycolysis splits one glucose molecule into two three-carbon pyruvate molecules. It releases energy, and some of that energy is captured in ATP. The pyruvate then enters the mitochondria, where the Krebs cycle, also called the citric acid cycle, finishes breaking down the glucose and loads electrons onto carrier molecules.

The last stage is oxidative phosphorylation. In this step, electrons move through an electron transport system in the inner mitochondrial membrane. This helps build a hydrogen ion gradient, and cells use that gradient to make most of the ATP in aerobic respiration.

Some organisms live where oxygen is scarce or where other chemicals dominate the environment. Extremophiles, which are bacteria found in places such as deep ocean hydrothermal vents or underwater caves, may use inorganic acceptors like nitrate or sulfur instead of oxygen. This is called anaerobic respiration.

When oxygen is absent and cellular respiration cannot continue, cells can use fermentation. Fermentation begins with glycolysis and makes some ATP, but it does not include oxidative phosphorylation, so it produces fewer ATP molecules than aerobic respiration. In humans, lactic acid fermentation occurs in red blood cells, which lack mitochondria, and in muscles during strenuous activity. Some bacteria also use lactic acid fermentation, which is why they are used to make yogurt.

Yeast carry out alcoholic fermentation. This pathway produces ethanol and carbon dioxide as byproducts. People use it to ferment beverages and to help dough rise.

Cellular respiration and photosynthesis are linked processes that move energy and matter through ecosystems. They are direct opposites in many ways because the products of one are the reactants of the other. Photosynthesis uses sunlight to build one glucose molecule from six carbon dioxide and six water molecules and releases six oxygen molecules. Cellular respiration uses six oxygen molecules to break one glucose molecule into six carbon dioxide and six water molecules while capturing energy as ATP and heat.

Scientists can measure respiration rates with a respirometer by tracking oxygen exchange. This method depends on the Ideal Gas Law, which relates pressure, volume, temperature, and the number of gas molecules. Respirometers contain potassium hydroxide, which traps carbon dioxide as potassium carbonate. As cells use oxygen, gas volume in the system drops, and scientists can use the ideal gas equation to calculate oxygen use.

Cellular respiration matters for health because mitochondria are essential to the process. Mitochondrial myopathies are neuromuscular diseases caused by mitochondrial damage, and they mainly affect nerve and muscle cells that need a lot of energy. Many poisons also block respiration. Cyanide, for example, inhibits ATP production through oxidative phosphorylation, and some antibiotics, chemotherapeutics, statins, and anesthetics can also interfere with mitochondrial function.
