Cell Structure

La vita, dagli organismi più grandi a quelli più minuscoli, è composta da cellule. Le cellule sono i mattoni della vita e forniscono i materiali necessari per l'esistenza della vita. 
Mentre le cellule di tutti i ceti sociali contengono strutture simili, come il DNA e le proteine, ci sono due diversi tipi di cellule in base a come tali strutture sono organizzate internamente. Le cellule procariotiche, quelle dei batteri e degli archei, sono semplici nella loro organizzazione. Ad esempio, mentre il loro genoma è densamente impacchettato, non è circondato da una membrana. Le cellule eucariotiche, quelle che compongono gli animali, le piante e i funghi sono più complesse. Compartimentalizzano le loro strutture interne in organelli, strutture legate alla membrana che svolgono funzioni specifiche. A differenza dei procarioti, il DNA eucariotico è racchiuso in una membrana nucleare. Altri organelli includono il reticolo endoplasmatico, il mitocondrio e il lisosoma. 
Le prime cellule sulla terra appaiono nella documentazione fossile di circa 3,7 miliardi di anni fa e si pensa che fossero procariotiche. Le cellule eucariotiche sono apparse per la prima volta circa 2,4 miliardi di anni fa, quando una cellula simile a un'ameba ha inghiottito una cellula batterica e ha formato una coesistenza stabile. Nel corso del tempo, questa cellula all'interno della cellula si è evoluta per formare strutture come il cloroplasto e il mitocondrio. 
Le piante e gli animali hanno anche esigenze metaboliche e strutturali diverse, il che porta a ulteriori differenze nelle loro cellule. Poiché le piante hanno bisogno di produrre la propria energia, le cellule vegetali contengono cloroplasti che aiutano la fotosintesi. La cellula vegetale include anche un grande vacuolo per immagazzinare proteine, metaboliti e acqua. Infine, le cellule vegetali hanno anche una parete cellulare rigida, necessaria per la crescita e la ritenzione idrica. 
Poiché le cellule sono così piccole, possono essere viste solo al microscopio. Uno dei modi più semplici per visualizzare le cellule è attraverso il microscopio a campo chiaro. Questo microscopio contiene una sorgente luminosa, una lente a condensatore per focalizzare la luce, una lente oculare per visualizzare il campione e una lente dell'obiettivo per l'ingrandimento. Un microscopio non è l'unica cosa necessaria per visualizzare l'invisibile. Le strutture all'interno delle cellule sono trasparenti, quindi le cellule devono essere colorate con coloranti. Ad esempio, la safranina viene utilizzata per colorare di rosso i nuclei delle cellule.

Le cellule rappresentano le unità biologiche più basilari di tutti gli organismi, sia che si tratti di organismi semplici e unicellulari come i batteri, sia che si tratti di grandi organismi multicellulari come gli elefanti e le sequoie giganti. A metà del XIX secolo, la Teoria delle Cellule è stata proposta per definire una cellula, che afferma:
<ul>
	<li>Ogni organismo vivente è costituito da una o più cellule.</li>
	<li>Le cellule sono le unità funzionali di tutti gli organismi.</li>
	<li>Tutte le cellule derivano da cellule preesistenti.</li>
</ul>
Tutte le cellule condividono caratteristiche comuni come avere una membrana plasmatica, un citoplasma, DNA e ribosomi. Una membrana plasmatica è un doppio strato fosfolipidico che circonda la cellula. Questo strato sottile e fluido attorno alle cellule serve a isolare il contenuto della cellula dal suo ambiente e regola lo scambio di materiale con il suo ambiente, facilitando anche le interazioni con altre cellule. All'interno della membrana plasmatica, la cellula è riempita con un fluido gelatinoso chiamato citoplasma che contiene molecole organiche, sali e altri materiali vitali per le funzioni della cellula. Pertanto, all'interno del citoplasma avvengono le reazioni biochimiche che supportano la vita, note come processi metabolici. I tipi di processi metabolici che una cellula può eseguire dipendono dalle sue informazioni genetiche. Tutte le cellule utilizzano il DNA come materiale genetico, che è il modello ereditario per costruire strutture e prodotti cellulari. Infine, tutte le cellule utilizzano i ribosomi per sintetizzare i loro prodotti proteici.
Esistono due tipi di cellule in base alla loro posizione nel materiale genetico: procariotiche, che significa "prima del nucleo", ed eucariotiche, che significano "vero nucleo". Pertanto, mentre entrambi i tipi di organismi hanno DNA, i procarioti come i batteri hanno nucleoidi, o componenti "simili a nuclei" invece di un nucleo, mentre gli eucarioti possiedono veri nuclei legati alla membrana per contenere il loro DNA. Inoltre, i procarioti sono relativamente piccoli, circa 0,1-5,0 micrometri (μm), rispetto agli eucarioti che possono tipicamente variare da 10-100 μm di dimensioni. Le piccole dimensioni dei procarioti consentono una distribuzione rapida e senza sforzo dei materiali all'interno della cellula e l'esecuzione dei processi metabolici, nonché una rapida rimozione di rifiuti o altri prodotti dalla cellula. Pertanto, le cellule eucariotiche possiedono strutture specializzate note come organelli, come i mitocondri o l'apparato di Golgi, per consentire l'esecuzione delle funzioni vitali.
<h4>La cellula eucariotica</h4>
La cellula eucariotica è un tratto derivato condiviso di tutti gli eucarioti, il che significa che ha avuto un'unica origine che da allora è stata ereditata da tutti gli eucarioti. Le prime cellule eucariotiche sono state osservate nei fossili circa 2,4 miliardi di anni fa e sono riconoscibili perché sono più grandidelle cellule procarioti1. L'origine di questo tipo di cellule è derivata da un evento endosimbiotico in cui una cellula simile a un'ameba ha inghiottito un batterio micrococcico e ha formato una coesistenza stabile2. I batteri inghiottiti si sono evoluti nei primi organelli produttori di energia, i mitocondri, che sono gli organelli del metabolismo aerobico nella cellula. I mitocondri hanno il loro genoma separato e sono di dimensioni simili ai procarioti. Contengono due strati di membrane che racchiudono due compartimenti distinti. Alcune delle reazioni che scompongono le biomolecole ad alta energia avvengono nel compartimento interno, mentre il compartimento esterno ospita le reazioni che catturano l'energia rilasciata da questi composti in molecole di adenosina trifosfato (ATP) da utilizzare come valuta energetica della cellula.
I nuclei e i mitocondri non sono le uniche strutture condivise dalle cellule eucariotiche. Altri organelli eucariotici ubiquitari sono il reticolo endoplasmatico liscio e ruvido (ER), l'apparato di Golgi, i lisosomi e i vacuoli. Reticolo endoplasmatico significa semplicemente "rete all'interno del plasma" e, come indica il nome, è una grande rete di membrane all'interno della cellula, in particolare intorno al nucleo. Parti dell'ER ruvido si estendono dalla membrana nucleare e si distinguono dall'ER liscio per il loro aspetto ruvido dovuto ai numerosi ribosomi sulla loro superficie. L'ER ruvido è il sito per la sintesi proteica, come le proteine che sono incorporate nella membrana plasmatica o le proteine che sono secrete dalla cellula. Al contrario, l'ER liscio produce prodotti a base di lipidi ma contiene anche enzimi per disintossicare le sostanze chimiche dannose. Quindi le cellule epatiche contengono abbondante ER liscio. Inoltre, le cellule muscolari contengono quantità significative di ER liscio a causa della funzione di immagazzinamento del calcio di questo organello, che è essenziale per la contrazione muscolare. L'apparato di Golgi seleziona, modifica e confeziona i prodotti cellulari all'interno di vescicole, che si fondono con la membrana plasmatica per rilasciare i prodotti. Alcune delle proteine prodotte nel reticolo endoplasmatico grezzo sono enzimi digestivi intracellulari. Questi enzimi sono impacchettati nell'apparato di Golgi in speciali vescicole chiamate lisosomi. La funzione principale dei lisosomi è quella di digerire le particelle di cibo inghiottite dalla cellula e le vecchie parti cellulari. I vacuoli sono sacche di membrana cellulare che fungono da unità di stoccaggio all'interno delle cellule. Possono servire a immagazzinare acqua per regolare il contenuto di acqua della cellula e a immagazzinare prodotti metabolici o persino molecole velenose, a seconda del tipo di cellula e dell'organismo.
<h4>Organelli specifici del regno</h4>
Le cellule eucariotiche hanno anche sviluppato organelli distinti, specifici per ogni regno. Ad esempio, il regno Plantae e Animalia sono entrambi eucarioti, tuttavia gli organelli delle cellule vegetali e animali differiscono in modi chiave che consentono loro di svolgere la loro vita rispettivamente come produttori e consumatori. Le piante terrestri devono crescere alte e avere steli rigidi per contenere le foglie, che usano per la fotosintesi. Devono anche essere in grado di trattenere l'acqua assorbita dalle radici. Le loro celle riflettono queste esigenze specifiche. A differenza delle cellule animali, le cellule vegetali hanno cloroplasti, che vengono utilizzati per la fotosintesi e spesso contengono il pigmento verde clorofilla. Inoltre, sono circondati da pareti cellulari, che sono strati esterni rigidi fatti di cellulosa per supportare la crescita e la ritenzione idrica. Poiché hanno bisogno di immagazzinare grandi quantità di acqua per mantenere la pressione dell'acqua nella cellula, hanno vacuoli più grandi rispetto alle cellule animali. Inoltre, le cellule vegetali hanno anche un altro tipo di organelli di stoccaggio specializzati chiamati plastidi, che contengono pigmenti e prodotti fotosintetici come l'amido. Queste differenze sono evidenti e distinguono le cellule vegetali da quelle animali: le cellule vegetali hanno generalmente una forma regolare e rettangolare a causa delle loro pareti cellulari rigide, mentre le cellule animali sono arrotondate e più irregolari.
<h4>Microscopia</h4>
Alcune cellule, come gli ovociti di rana, sono abbastanza grandi da essere viste ad occhio nudo, ma la maggior parte delle cellule non può essere vista senza alcun aiuto visivo. Pertanto, gli scienziati utilizzano tecniche di microscopia per studiare le strutture cellulari e distinguere i tipi di cellule l'una dall'altra. Mentre i microscopi sono in grado di ingrandire oggetti che sono difficili o impossibili da vedere con l'occhio umano, la maggior parte dei tessuti manca naturalmente di pigmentazione. Pertanto, sono state create soluzioni in grado di colorare selettivamente le cellule in base alla loro composizione molecolare. Ciò consente ai ricercatori di distinguere tra organelli in una cellula, tipi di tessuto in uno stelo di pianta e strati di grasso negli animali, solo per citare alcuni esempi. Il colorante blu di metilene colora gli acidi nucleici delle cellule morte, legandosi al DNA caricato negativamente. La soluzione di safranina è un altro colorante biologico che colora di rosso i nuclei cellulari. Le celle devono essere immerse nelle soluzioni coloranti solo per un breve periodo di tempo e possono essere montate immediatamente dopo la fase di colorazione. Le tecniche di montaggio comunemente utilizzate sono il montaggio a umido e l'immersione in olio. Una montatura bagnata viene creata raccogliendo un campione e mettendolo su un vetrino con del liquido tra il vetrino e il vetrino. I campioni cellulari sono sospesi in liquidi come acqua o glicerolo. Il glicerolo è meglio usarlo con colture vive, perché impedisce la proliferazione dei batteri3. L'olio da immersione può essere aggiunto sulla parte superiore del vetrino coprioggetti per migliorare la visualizzazione del campione ad alto ingrandimento. Ciò si ottiene perché l'olio ha lo stesso indice di rifrazione del vetro, il che significa che consente alla luce di attraversarlo così come farebbe il vetro. L'interfaccia vetro-aria diffonde la luce più dell'olio o del vetro, quindi la nitidezza dell'immagine è influenzata quando i campioni sono montati "a secco" o senza olio. Una volta che le cellule sono state colorate e montate, sono pronte per essere studiate al microscopio.
Esistono varie tecniche di microscopia, dalla tecnologia di scansione elettronica che ha permesso ai ricercatori di visualizzare oggetti a livello atomico all'imaging fluorescente di cellule vive che consente il monitoraggio in tempo reale del movimento delle molecole all'interno delle singole cellule4. La microscopia a campo chiaro è la tecnica di microscopia più semplice, che richiede solo una sorgente di luce alogena, una lente a condensatore per focalizzare la luce, una lente oculare per visualizzare l'immagine e una lente dell'obiettivo per ingrandire l'immagine. Con qualsiasi tecnica di microscopia, è importante comprendere le parti di un microscopio prima di utilizzarne uno. In generale, i microscopi composti utilizzati per l'imaging in campo chiaro hanno un oculare nella parte superiore del cannocchiale, che è attaccato alla testa e agli obiettivi. L'oculare ha un ingrandimento di 10X e le lenti dell'obiettivo sono impostate su un ingrandimento particolare in un intervallo da 4X-100X. Ci sono da tre a cinque obiettivi su un microscopio standard. Gli obiettivi puntano verso il basso, che è il luogo in cui viene posizionato un campione per la visualizzazione. Il palco è spesso dotato di parti meccaniche e clip per tenere una diapositiva e spostarla durante la visualizzazione. Un'apertura è un foro nel palco attraverso il quale la luce può passare. Questa luce è controllata da una lente a condensatore regolabile sopra un illuminatore o una sorgente luminosa. Per controllare lo zoom del tavolino per l'oggetto da osservare, i microscopi sono dotati di manopole di regolazione della messa a fuoco grossolana e fine. La manopola di messa a fuoco grossolana si muove su una scala più ampia rispetto alla messa a fuoco fine, ma si trovano sullo stesso asse. La messa a fuoco fine è utile quando l'oggetto sul palco è stato avvicinato agli obiettivi. È importante non lasciare che la lente dell'obiettivo tocchi l'oggetto sul tavolino, poiché può graffiare la lente. Gli oggetti devono sempre essere visualizzati prima sull'obiettivo a ingrandimento più basso e messi a fuoco in modo chiaro prima di passare a obiettivi a ingrandimento più elevato.
La microscopia è uno strumento importante per molti aspetti del campo medico, tra cui la ricerca, la diagnosi e il trattamento. Questo ha l'applicazione dell'uso della nanotecnologia in medicina, come nuovo metodo di trattamento al posto di una chirurgia più invasiva5. I chirurghi fanno anche uso di microscopi, alcuni dei quali sono stati modificati per essere montati sulla testa di un chirurgo e vengono azionati con pedali. Questi sono di ingrandimento molto inferiore anche rispetto ai microscopi ottici utilizzati oggi, ma facilitano l'esecuzione sicura di procedure delicate, come l'ottica e la neurochirurgia.

Cells are the basic units of life, and their structures help explain how living things work. Cell theory says that every living organism is made of one or more cells, cells are the functional units of organisms, and new cells come from preexisting cells. Cells also share key features, including a plasma membrane, cytoplasm, DNA, and ribosomes.

The plasma membrane is a phospholipid bilayer that surrounds the cell. It separates the cell from its environment and controls what enters and leaves. Inside it, the cytoplasm is a gel-like fluid that contains organic molecules, salts, and other materials needed for cell activity. Many metabolic processes, or life-supporting chemical reactions, take place there.

Cells use DNA as their genetic material, which acts as the blueprint for cellular structures and products. Ribosomes make proteins. Cells are often grouped as prokaryotic or eukaryotic based on where their genetic material is found. Prokaryotes, such as bacteria, have nucleoids instead of a true nucleus, while eukaryotes have membrane-bound nuclei.

Prokaryotic cells are usually small, about 0.1 to 5.0 micrometers, while eukaryotic cells are often 10 to 100 micrometers. Their small size helps prokaryotes move materials quickly and remove waste efficiently. Eukaryotic cells use organelles, such as mitochondria and the Golgi apparatus, to carry out important functions.

Eukaryotic cells are thought to have a shared origin and appear in fossils about 2.4 billion years ago. The earliest eukaryotic cells were larger than prokaryotic cells and likely formed through an endosymbiotic event. In this event, an amoeba-like cell engulfed a micrococcal bacterium, and that bacterium later became the mitochondrion. Mitochondria perform aerobic metabolism and have their own genome.

Mitochondria have two membranes that enclose two compartments. Some reactions that break down high-energy biomolecules happen in the inner compartment. Other reactions in the outer compartment capture released energy and convert it into ATP, the cell’s energy currency. Eukaryotic cells also contain the smooth and rough endoplasmic reticulum, the Golgi apparatus, lysosomes, and vacuoles.

The endoplasmic reticulum means “network inside the plasma,” and it forms a membrane network around the nucleus. Rough ER has ribosomes on its surface and makes proteins, including proteins for the plasma membrane and proteins that are secreted from the cell. Smooth ER makes lipid-based products and helps detoxify harmful chemicals, which is why liver cells contain much of it. Muscle cells also have a lot of smooth ER because it stores calcium needed for contraction.

The Golgi apparatus sorts, modifies, and packages cellular products into vesicles. Some proteins made in the rough ER are digestive enzymes. The Golgi packages these enzymes into lysosomes, which digest food particles taken into the cell and break down old cell parts. Vacuoles are membrane sacs used for storage, including water, metabolic products, or even poisonous molecules, depending on the cell.

Plant and animal cells are both eukaryotic, but they do not have the same organelles. Plant cells have chloroplasts for photosynthesis and often contain chlorophyll, the green pigment. They also have cell walls made of cellulose, which give support and help with water retention. Their larger vacuoles help maintain water pressure, and they also contain plastids that store pigments and products such as starch.

These differences make plant cells and animal cells easy to tell apart. Plant cells usually have a regular, rectangular shape because of their rigid cell walls. Animal cells are more rounded and irregular. Cell walls, chloroplasts, and large vacuoles are key features that distinguish plant cells from animal cells.

Many cells are too small to see without help, so scientists use microscopy to study them. Some cells, such as frog oocytes, are large enough to see with the naked eye, but most samples need magnification. Because many tissues lack color, stains are used to highlight cell parts and tissue types. Methylene blue stains nucleic acids in dead cells by binding to DNA, and safranin stains cell nuclei red.

After staining, cells can be mounted as a wet mount or with immersion oil. In a wet mount, the sample is placed on a slide with liquid between the slide and coverslip. Water or glycerol can be used as the liquid, and glycerol is better for live cultures because it prevents bacterial growth. Immersion oil improves high-magnification viewing because it has the same refractive index as glass and reduces light scattering.

Once stained and mounted, the sample is ready for the microscope. Microscopy methods range from electron scanning technology, which can view objects at atomic scale, to fluorescent live-cell imaging, which tracks molecules in real time. Bright-field microscopy is the simplest method and uses a halogen light source, condenser lens, ocular lens, and objective lens.

Compound microscopes have an eyepiece, a head with objectives, and a stage for the specimen. The eyepiece usually has 10X magnification, and objective lenses commonly range from 4X to 100X. The stage may have clips and mechanical parts to move the slide, and light passes through an aperture in the stage. Coarse and fine focus knobs control sharpness, and the slide should be viewed first at low magnification before moving higher.

Microscopy is important in medicine for research, diagnosis, and treatment. It also supports newer approaches such as nanotechnology in medicine, which can reduce the need for more invasive surgery. Surgeons use modified microscopes as well, including head-mounted models operated with foot pedals. These lower-magnification tools help doctors perform delicate procedures such as optical and neurosurgery safely.
