La replicazione del DNA avviene sintetizzando nuovi filamenti di DNA usando filamenti esistenti come modello. Le due nuove doppie eliche contengono ciascuna un filamento modello originale e un filamento-figlio di nuova sintesi, motivo per cui questo processo è conosciuto come replicazione semiconservativa. Per iniziare la replicazione, un enzima, l'elicasi, apre l'elica del DNA e rompe i legami idrogeno fra i due filamenti.

Quindi separa i singoli filamenti, formando una struttura a forma di Y, detta forcella di replicazione, in cui è possibile accedere ai filamenti modello da ulteriori enzimi. Un altro enzima, la primasi, aggiunge brevi frammenti di RNA, detti primer, su ciascun filamento del modello. Questi primer sono essenziali per la sintesi del DNA, poiché la DNA polimerasi può solo aggiungere nucleotidi a un filamento esistente.

La DNA polimerasi si aggiunge ai filamenti-figli su entrambi i filamenti del DNA modello. L'aggiunta di nucleotidi è guidata dalla sequenza del filamento di DNA originale secondo le regole di accoppiamento del DNA. La sintesi di uno dei filamenti-figli, il filamento principale, avviene nella direzione del movimento della forcella di replicazione.

L'altro filamento, il filamento ritardatario, viene sintetizzato nella direzione opposta. Questo porta al filamento principale che viene sintetizzato come polimero continuo, mentre il filamento ritardatario è sintetizzato come frammenti brevi. Questo si verifica perché il DNA può essere sintetizzato solo nella direzione da 5'a 3'Prima della loro aggiunta al polimero in crescita, i nucleotidi esistono come desossiribonucleosi di trifosfati, con tre fosfati attaccati al quinto carbonio sullo zucchero.

Questo nucleotide libero trifosfato reagisce con i gruppi idrossilici 3'Questo è l'OH che è attaccato al terzo carbonio dello zucchero alla fine del filamento in crescita. La reazione provoca il rilascio di pirofosfato e la formazione di un legame fosfodiestere fra i due nucleotidi. Dopo la sintesi dei nuovi filamenti, RNase H o varianti aggiuntive della DNA polimerasi, rimuovete i primer e sintetizzate il DNA al loro posto.

Gli spazi fra i frammenti vengono dunque sigillati dalla DNA ligasi per generare un filamento continuo. L'aggiunta di nucleotidi continua fino a quando due forcelle di replicazione si incontrano, determinando la completa replicazione.

DNA Replication

La replicazione del DNA comporta la separazione dei due filamenti della doppia elica, ciascun filamento funge da modello da cui viene copiato il nuovo filamento complementare. Dopo la replicazione, ciascun DNA a doppio filamento include un filamento parentale o “vecchio” e un filamento “nuovo”. Questo è noto come replicazione semiconservativa. Le molecole di DNA risultanti hanno la stessa sequenza e sono divise equamente nelle due cellule figlie.

Replicazione nei Procarioti

La replicazione del DNA utilizza un gran numero di proteine ed enzimi. L'enzima elicasi separa i due filamenti di DNA. Mentre l’elicasi si muove lungo il DNA, separa i due filamenti per formare una struttura a forma di Y conosciuta come forca di replicazione. Successivamente, l'enzima primasi aggiunge brevi tratti di RNA noti come primer per avviare la sintesi del DNA da parte della DNA polimerasi, l'enzima responsabile della sintesi del DNA. Nei batteri sono noti tre tipi principali di DNA polimerasi: DNA pol I, DNA pol II e DNA pol III. DNA pol III è l'enzima necessario per la sintesi del DNA; DNA pol I e DNA pol II sono necessari principalmente per la riparazione. Il DNA pol III aggiunge desossiribonucleotidi, ciascuno complementare a un nucleotide sul filamento modello, uno per uno al gruppo 3'-OH della catena di DNA in crescita. La DNA polimerasi III può estendersi solo nella direzione da 5' a 3'. L'aggiunta di questi nucleotidi richiede energia. Questa energia è presente nei legami di tre gruppi fosfato attaccati a ciascun nucleotide, in modo simile a come l'energia viene immagazzinata nei legami fosfato dell'adenosina trifosfato (ATP). Quando il legame tra i fosfati viene rotto e viene rilasciato il difosfato, l'energia rilasciata consente la formazione di un legame fosfodiesterico covalente mediante sintesi di disidratazione.

La doppia elica del DNA è antiparallela; cioè, un filo è orientato nella direzione da 5' a 3' e l'altro è orientato nella direzione da 3' a 5'. Durante la replicazione, un filamento, che è complementare al filamento di DNA parentale da 3' a 5', viene sintetizzato continuamente verso la forca di replicazione poiché la polimerasi può aggiungere nucleotidi in questa direzione. Questo filamento sintetizzato continuamente è noto come filamento principale. L'altro filamento, complementare al DNA parentale da 5' a 3', cresce lontano dalla forca di replicazione, quindi la polimerasi deve tornare indietro verso la forca di replicazione per iniziare ad aggiungere basi a un nuovo primer, sempre nella direzione opposta alla forca di replicazione. . Lo fa finché non incontra il filamento precedentemente sintetizzato, quindi torna indietro. Questi passaggi producono piccoli frammenti di sequenze di DNA noti come frammenti di Okazaki, ciascuno separato da un primer di RNA. Il filamento con i frammenti di Okazaki è noto come filamento ritardato e si dice che la sua sintesi sia discontinua.

Dopo la sintesi da parte della DNA polimerasi III, i primer vengono rimossi dall'attività di esonucleasi della DNA polimerasi I e gli spazi vuoti vengono riempiti. Le intaccature che rimangono tra il DNA appena sintetizzato (che ha sostituito il primer dell'RNA) e il DNA precedentemente sintetizzato vengono sigillato dall'enzima DNA ligasi che catalizza la formazione di un legame covalente fosfodiestere tra l'estremità 3'-OH di un frammento di DNA e l'estremità 5' fosfato dell'altro frammento, stabilizzando lo scheletro zucchero-fosfato della molecola di DNA.

Replicazione negli eucarioti

I genomi eucariotici sono molto più complessi e più grandi dei genomi procariotici e sono tipicamente composti da più cromosomi lineari. Il genoma umano, ad esempio, ha 3 miliardi di paia di basi per set aploide di cromosomi e durante la replicazione vengono inseriti 6 miliardi di paia di basi. La velocità di replicazione è di circa 100 nucleotidi al secondo - 10 volte più lenta della replicazione procariotica.

Le fasi essenziali della replicazione negli eucarioti sono le stesse dei procarioti. Prima che la replicazione possa iniziare, il DNA deve essere reso disponibile come modello. Il DNA eucariotico è altamente avvolto e impacchettato, il che è favorito da molte proteine, compresi gli istoni. Dopo l'inizio della replicazione, in un processo simile a quello riscontrato nei procarioti, l'allungamento è facilitato dalle DNA polimerasi eucariotiche. Il filamento principale è continuamente sintetizzato dall'enzima polimerasi eucariotica pol δ, mentre il filamento ritardato è sintetizzato da pol ε. L'enzima ribonucleasi H (RNasi H), invece della DNA polimerasi come nei batteri, rimuove il primer dell'RNA, che viene poi sostituito con nucleotidi del DNA. Gli spazi rimasti vengono sigillati dalla DNA ligasi.

Come nei procarioti, la DNA polimerasi eucariotica può aggiungere nucleotidi solo nella direzione da 5’ a 3’. Nel filamento principale, la sintesi continua finché non raggiunge l'estremità del cromosoma o un'altra forcella replicativa che procede nella direzione opposta. Sul filamento ritardato, il DNA viene sintetizzato in brevi tratti, ciascuno dei quali viene avviato da un primer separato. Quando la forca replicativa raggiunge la fine del cromosoma lineare, non esiste un posto dove creare un primer per il frammento di DNA da copiare all'estremità del cromosoma. Queste estremità rimangono quindi spaiate e, nel tempo, possono accorciarsi progressivamente man mano che le cellule continuano a dividersi.

DNA replication copies genetic material before a cell divides. Each original DNA strand serves as a template for a new complementary strand. The result is semiconservative replication, which means each new DNA molecule contains one old strand and one new strand. The two daughter cells then receive DNA with the same sequence.

Replication starts when helicase separates the two strands of the double helix. This creates a Y-shaped replication fork. Primase then adds short RNA primers that give DNA polymerase a place to begin DNA synthesis. In bacteria, the main DNA polymerases are DNA pol I, DNA pol II, and DNA pol III. DNA pol III carries out DNA synthesis, while DNA pol I and DNA pol II mainly help with repair.

DNA pol III adds deoxyribonucleotides one at a time to the 3'-OH end of the growing strand. It can extend DNA only in the 5' to 3' direction. The energy for this process comes from the three phosphate groups on each nucleotide. When a phosphate bond is broken and diphosphate is released, that energy helps form the phosphodiester bond in the DNA backbone.

Because the DNA double helix is antiparallel, the two template strands face opposite directions. The strand copied toward the replication fork is made continuously and is called the leading strand. The other strand is copied away from the fork. It must start over with new primers as the fork opens, so it is made in short stretches called Okazaki fragments. This strand is the lagging strand, and its synthesis is discontinuous.

After DNA pol III finishes, DNA pol I removes the RNA primers in bacteria, and the gaps are filled in. DNA ligase then seals the remaining nicks by forming covalent phosphodiester bonds between adjacent DNA fragments. This step strengthens the sugar-phosphate backbone of the new DNA molecule.

Eukaryotic DNA replication follows the same basic steps, but the genome is larger and more complex. Eukaryotic chromosomes are linear and have many origins of replication. In humans, the genome has 3 billion base pairs per haploid set, and about 6 billion base pairs are copied during replication. Human chromosomes also have about 30,000 to 50,000 origins of replication, and replication occurs at about 100 nucleotides per second, which is much slower than in prokaryotes.

Before replication begins, eukaryotic DNA must be made available as a template. It is tightly packed and supercoiled, and histones help organize this packaging. After initiation, eukaryotic DNA polymerases carry out elongation. Polymerase delta (pol δ) synthesizes the leading strand, and polymerase epsilon (pol ε) synthesizes the lagging strand. RNase H removes the RNA primers, DNA nucleotides replace them, and DNA ligase seals the final gaps. Because eukaryotic DNA polymerase also works only in the 5' to 3' direction, the lagging strand is still made in short pieces. Near the ends of linear chromosomes, there is no place to add a final primer, so these ends remain unpaired and can become shorter over time as cells keep dividing.
