Introduction to Enzymes

Gli enzimi sono catalizzatori biologici che accelerano la velocità di reazione senza essere consumati. Sono tipicamente proteine situate nel citoplasma, all'interno degli organelli e nelle membrane cellulari o degli organelli.
Gli enzimi possono funzionare a livello intracellulare o possono essere secreti extracellulare. La maggior parte degli enzimi richiede una temperatura e un pH specifici per funzionare alla massima efficienza.
I substrati sono i reagenti che si legano specificamente ai siti attivi, piccole regioni dell'enzima in cui avviene la reazione. Le parti rimanenti dell'enzima forniscono struttura all'enzima o interagiscono con altre molecole per promuovere o inibire la reazione.
Gli enzimi cambiano la loro forma per legare i loro substrati e catalizzare la conversione dei substrati in prodotti. Dopo la reazione, l'enzima rilascia il prodotto e ritorna alla sua conformazione originale, che consente all'enzima di catalizzare ulteriori cicli della reazione.

L'uso degli enzimi da parte dell'uomo risale al 7000 a.C. Gli esseri umani hanno utilizzato per la prima volta gli enzimi per fermentare gli zuccheri e produrre alcol senza sapere che si trattava di una reazione catalizzata da enzimi. Wilhelm Kuhne coniò il termine "enzima" nel 1877 dalle parole greche "en" che significa "dentro" o "dentro" e "zyme" che significa "lievito".

La maggior parte degli enzimi sono proteine che accelerano le reazioni biochimiche senza essere consumate. Gli enzimi contengono uno o più siti attivi che legano i substrati e li convertono in prodotti. Gli inibitori sono molecole che fermano una reazione catalizzata da un enzima. Gli inibitori competitivi sono simili alle molecole del substrato e competono con esse per legarsi ai siti attivi. Al contrario, gli inibitori non competitivi si legano ai siti allosterici e modificano la conformazione dell’enzima per ridurre il legame con il substrato.

Enzymes are proteins that speed up biochemical reactions without being used up. Humans have used enzymes since about 7000 BCE, when they fermented sugars to make alcohol without knowing the reaction was enzyme-catalyzed. The word enzyme was coined by Wilhelm Kuhne in 1877. It comes from the Greek words en, meaning “in” or “within,” and zyme, meaning “yeast.”

Enzymes work through active sites, which are regions that bind substrates and help convert them into products. Many enzymes also have allosteric sites. These are extra binding sites where molecules other than substrates can attach and either activate or inhibit the enzyme.

Some enzymes need cofactors to carry out catalysis. Cofactors are metal ions or organic molecules that bind to the active site and help the reaction happen. An enzyme without its cofactor is called an apoenzyme. When the cofactor is attached, it becomes a holoenzyme. An organic cofactor is called a coenzyme, and most coenzymes are derived from vitamins.

Enzyme activity can also be blocked by inhibitors. Competitive inhibitors are similar to the substrate and compete for the active site. Non-competitive inhibitors bind to allosteric sites instead. This changes the enzyme’s shape and reduces substrate binding.
