Le ammine secondarie, tra cui alchile e arile, dopo il trattamento con NaNO2 e un forte acido producono N-nitrosammine secondarie.
Ricordiamo che una soluzione acidificata di NaNO2 genera HNO2. L'instabile HNO2 subisce inoltre la protonazione e la successiva disidratazione per formare lo ione nitrosonio, un elettrofilo.
Il catione nitrosilico, generato in situ, viene attaccato dalle ammine secondarie nucleofile per formare uno ione N-nitrosammonio.
Lo ione, in seguito alla deprotonazione da parte di molecole di solvente, provoca la formazione di N-nitrosammine secondarie.
A differenza delle N-nitrosammine primarie, derivate dalle ammine primarie, le N-nitrosammine secondarie sono stabili perché l'assenza del protone N-H impedisce un ulteriore tautomerismo e l'eventuale formazione di diazonio.
Di solito si separano come liquidi oleosi dalla miscela di reazione.
In generale, le N-nitrosammine secondarie hanno rare applicazioni sintetiche. Tuttavia, molti di essi sono noti agenti cancerogeni presenti negli alimenti trasformati o nel tabacco.

Le ammine secondarie reagiscono con l'acido nitroso per formare N-nitrosammine, come illustrato in Figura 1. L'acido nitroso, un acido debole e instabile, si forma in situ da una soluzione acquosa di nitrito di sodio e acidi forti, come acido cloridrico o acido solforico, in condizioni di freddo. In presenza di un acido, l'acido nitroso viene protonato. La successiva perdita di acqua porta alla formazione dell'elettrofilo noto come ione nitrosonio.

L'ammina secondaria che agisce come nucleofilo reagisce con lo ione nitrosonio per dare un altro ione noto come ione N-nitrosammonio. Lo ione risultante viene quindi deprotonato dall'acqua per formare N-nitrosammine secondarie. Questo meccanismo di reazione è illustrato in Figura 2. Di solito, le N-nitrosammine secondarie sono meno significative per l'uso commerciale, tuttavia, sono studiate a causa delle loro potenti proprietà cancerogene. In Norvegia (1962), un'epidemia di intossicazione alimentare nelle pecore fu attribuita alla farina di pesce trattata con nitriti. Secondary amines can form N-nitrosamines when they react with nitrous acid. Nitrous acid is weak and unstable, so it is made in the reaction mixture from sodium nitrite and a strong acid such as hydrochloric acid or sulfuric acid. The reaction is carried out in cold conditions.

In acid, nitrous acid becomes protonated. It then loses water and forms the nitrosonium ion, a reactive electrophile. The secondary amine acts as a nucleophile and attacks this ion. That step gives an N-nitrosammonium ion.

Water then removes a proton from the ion to make the secondary N-nitrosamine. These products can be separated from the mixture as oily yellow liquids. Although they are not usually important for commercial use, they are studied because many are potent carcinogens.

The transcript also links this chemistry to food safety. In 1962, a food-poisoning outbreak in sheep in Norway was traced to nitrite-treated fish meal. Sodium nitrite is used to preserve foods and meat, but in the stomach it can form nitrous acid. That acid can react with secondary amines in food to produce highly carcinogenic N-nitrosamines, so FDA limits sodium nitrite in meat preservation to 50 to 125 ppm. N-nitrosamines are also found in cigarette and tobacco smoke.
