Turnover Number and Catalytic Efficiency

Il numero di turnover o kcat indica la rapidità con cui un enzima può convertire le molecole del substrato in prodotti.
KCAT è uguale al numero massimo di molecole di substrato che possono essere trasformate in un dato tempo per sito attivo dell'enzima.
I numeri di turnover di diversi enzimi variano da meno di una molecola di substrato a milioni di molecole al secondo.
Per calcolare kcat, la velocità massima o Vmax di una reazione catalizzata da un enzima viene divisa per la concentrazione totale dell'enzima.

Il turnover number di un enzima è il numero massimo di molecole di substrato che può trasformare nell'unità di tempo. I numeri di turnover per la maggior parte degli enzimi vanno da 1 a 1000 molecole al secondo. La catalasi ha il numero di turnover più alto conosciuto, in grado di convertire fino a 2,8×106 molecole di perossido di idrogeno in acqua e ossigeno al secondo. Il lisozima ha il numero di turnover più basso conosciuto di mezza molecola al secondo.

La chimotripsina è un enzima pancreatico che scompone le proteine durante la digestione. Il numero di turnover della chimotripsina è di 100 molecole al secondo. Se questa reazione dovesse avvenire senza catalizzazione, i legami peptidici impiegherebbero centinaia di anni per rompersi nell’acqua a pH neutro. Pertanto, l'elevato numero di turnover della chimotripsina aiuta una rapida digestione delle proteine nell'intestino.

L'enzima ribulosio 1,5-bifosfato carbossilasi ossigenasi o RuBisCO ha un numero di turnover molto basso fissando 3 molecole di CO2 al secondo ed è uno degli enzimi più lenti. Tuttavia, l’abbondanza di RuBisCO in natura compensa il basso numero di turnover. RuBisCO costituisce circa il 50% delle proteine totali presenti nelle foglie.

Un enzima con un numero di turnover elevato potrebbe non essere necessariamente altamente efficiente. Enzymes can work at very different speeds, and their turnover number shows how fast they convert substrate molecules into products. Turnover number is the maximum number of substrate molecules an enzyme can transform per unit time. For most enzymes, this value is between 1 and 1000 molecules per second.

Some enzymes are especially fast. Catalase has the highest known turnover number and can convert up to 2.8×10^6 molecules of hydrogen peroxide into water and oxygen each second. Lysozyme has the lowest known turnover number, at half a molecule per second.

Digestion also depends on enzyme speed. Chymotrypsin is a pancreatic enzyme that breaks down proteins during digestion, and its turnover number is 100 molecules per second. Without an enzyme, peptide bonds would take hundreds of years to break in water at neutral pH. Chymotrypsin helps make protein digestion in the intestine much faster.

Not every slow enzyme is unimportant. RuBisCO, or ribulose 1,5-bisphosphate carboxylase oxygenase, has a very low turnover number and fixes only 3 molecules of CO2 per second. It is one of the slowest enzymes, but it is very abundant in nature. RuBisCO makes up around 50% of the total protein found in leaves.

A high turnover number does not always mean high catalytic efficiency. Catalytic efficiency is the ratio of turnover number, kcat, to affinity, KM. An enzyme also needs a low KM for its substrate to be efficient. The average catalytic efficiency of most enzymes is about 10^5 M^-1 s^-1, so they are moderately efficient. A few enzymes reach 10^8 to 10^9 M^-1 s^-1 and are called superefficient or catalytically perfect.
