Cationic Metabolites

Cationic metabolites are small biochemical compounds that carry a net positive charge, often because their amine groups are protonated under cellular conditions. Their charge depends on pH and molecular structure, influencing solubility, membrane transport, electrostatic interactions, and recognition by metabolic enzymes. Examples include protonated amino compounds and polyamines, which participate in biosynthetic and regulatory pathways. Characterizing cationic metabolites helps researchers interpret metabolomics data, investigate nutrient and ion transport, and assess changes in cellular metabolism associated with disease or environmental stress. Analytical methods that separate and detect charged molecules are therefore important for measuring their concentrations and biological roles.

Detection of Human Fecal Metabolites Using a Bacterial Biosensor

2026

Begin with a bacterial biosensor culture suspended in a medium. These engineered bacteria produce a transcription factor that, upon binding to a specific fecal metabolite, activates a promoter associated with a fluorescent reporter gene.To this culture, add a filtered human fecal sample containing metabolites, such as lactate, that are involved in host-microbiome interactions.Dilute this mixture with a fresh medium to maintain an optimal reaction volume.Incubate without shaking to mimic the...

We describe here the procedures for the extraction and purification of mRNA and metabolites from Drosophila heads. We are applying these techniques to better understand the cellular perturbations underlying neuronal degeneration. These methodologies can be easily scaled and adapted for other "omic" projects.

Begin with a bacterial suspension in nutrient media.Add the suspension to a multi-well plate and incubate. The nutrients in the media facilitate bacterial growth and attachment. The adhered bacteria secrete extracellular polymeric substances consisting of polysaccharides, proteins, and DNA to form a layer of biofilm.Wash multiple times with buffer to remove unbound bacteria.Invert the plate to ensure the removal of residual liquid from the well and allow the biofilm to dry.Treat with a fixative...
