Fluorescent Protein Quantification

Fluorescent protein quantification is the measurement of fluorescently tagged protein abundance or concentration by detecting the light it emits, providing a noninvasive readout of protein production and behavior. When excitation light reaches a fluorophore, it raises electrons to a higher energy state; as they return to the ground state, emitted fluorescence is measured and compared with calibrated standards under controlled conditions. In biochemistry, spectrofluorometers and microplate readers use this signal to evaluate recombinant protein expression, purification yield, and assay performance. Quantification also supports studies of protein localization, interactions, and dynamic changes in living or fixed cells.

The present protocol describes a procedure to perform fluorescent size exclusion chromatography (FSEC) on membrane proteins to assess their quality for downstream functional and structural analysis. Representative FSEC results collected for several G-protein coupled receptors (GPCRs) under detergent-solubilized and detergent-free conditions are presented.

Automated Quantification of Synaptic Fluorescence in C. elegans

The abundance of neurotransmitter receptors clustered at synapses strongly influences synaptic strength. This method quantifies fluorescently-labeled neurotransmitter receptors in three dimensions with single-synapse resolution in C. elegans, allowing hundreds of synapses to be rapidly characterized within a single sample without distortions introduced by z-plane projection.
