Biochip Reusability

Biochip reusability is the ability to clean, regenerate, and redeploy a biochip for multiple analytical or biological experiments while preserving its structural and functional performance. Reuse typically involves removing biological samples and contaminants from microfluidic channels or sensing surfaces through controlled washing, chemical treatment, or other decontamination steps, followed by verification that the chip retains reliable fluid handling and signal response. In bioengineering, reusable biochips can reduce material consumption, experimental costs, and waste while supporting repeated measurements and standardized workflows. Their practical value depends on maintaining assay accuracy, preventing carryover, and ensuring that regeneration does not damage sensitive biological interfaces.

Presented is the protocol for co-immobilizing whole-cell biocatalysts for cofactor regeneration and improved reusability, using the production of L-xylulose as an example. The cofactor regeneration is achieved by coupling two Escherichia coli strains expressing functionally complementary enzymes; the whole-cell biocatalyst immobilization is achieved by cell encapsulation in calcium alginate beads.

All-solid-state ion-selective electrodes (ASSISEs) constructed from a conductive polymer (CP) transducer provide several months of functional lifetime in liquid media. Here, we describe the fabrication and calibration process of ASSISEs in a lab-on-a-chip format. The ASSISE is demonstrated to have maintained a near-Nernstian slope profile after prolonged storage in complex biological media.
