Chemokine Sdf-1 Alpha

Chemokine SDF-1 alpha, also known as CXCL12, is a signaling protein that directs the movement, retention, and positioning of cells in developing and injured tissues. It binds primarily to the receptors CXCR4 and CXCR7, activating intracellular pathways that guide cell migration and influence survival, adhesion, and tissue homing. In bioengineering, researchers incorporate SDF-1 alpha into biomaterials, hydrogels, and engineered scaffolds to recruit stem or progenitor cells and regulate tissue regeneration. Controlled delivery can help create chemotactic gradients that improve vascularization, cell organization, and repair, making this chemokine important for regenerative medicine and cell-guiding strategies.

Preparation and Characterization of SDF-1α-Chitosan-Dextran Sulfate Nanoparticles

The objective of this protocol is to incorporate SDF-1α, a stem cell homing factor, into dextran sulfate-chitosan nanoparticles. The resultant particles are measured for their size and zeta potential, as well as the content, activity, and in vitro release rate of SDF-1α from the nanoparticles.

The three-dimensional flow chamber device is a novel in vitro technology for the quantitative and step-wise evaluation of the extravasation cascade of cells circulating under conditions of physiological shear stress. The device therefore fills a critical need for basic, preclinical, and clinical studies of cell migration.

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.
