Electric Cell Sensing

Electric cell sensing is a bioengineering approach that detects and monitors cell behavior through changes in electrical signals, often without fluorescent labels or destructive sampling. In electrode-based systems, an applied electrical field passes through or near cultured cells, and cell attachment, spreading, proliferation, migration, or changes in barrier integrity alter impedance, resistance, or conductance; instruments record these changes over time. This label-free, real-time method supports studies of cell adhesion, wound healing, epithelial and endothelial barriers, cytotoxicity, and drug responses. By converting cellular activity into quantitative electrical measurements, electric cell sensing helps connect dynamic cell physiology with engineered assays and therapeutic development.

This protocol reviews Electric Cell-substrate Impedance Sensing, a method to record and analyze the impedance spectrum of adherent cells for the quantification of cell attachment, proliferation, motility, and cellular responses to pharmacological and toxic stimuli. Detection of endothelial permeability and assessment of cell-cell and cell-substrate contacts are emphasized.
