Methylene Blue Redox Indicator

Methylene blue redox indicator is a reversible chemical sensor that signals changes in oxidation-reduction conditions through a visible color change. In its oxidized form, methylene blue appears blue; when it accepts electrons from reducing agents, it is converted to colorless leucomethylene blue, and exposure to oxygen can restore the blue form. This cycling provides a simple way to monitor electron transfer, oxygen availability, and reducing conditions in bioengineering experiments. Researchers use it in redox assays, microbial and cellular respiration studies, and demonstrations of biochemical energy conversion, where its clear visual response supports process monitoring and experimental interpretation.

(2021)This video demonstrates the imaging of mitochondrial redox state in primary neurons using a ratiometric fluorescent indicator. By applying two excitation wavelengths under a fluorescence microscope, the fluorescence intensity ratio is measured to assess mitochondrial oxidation. The protocol includes N-methyl-D-aspartate (NMDA) stimulation...

Oxidation and Reduction Some chemical reactions can be classified as reduction-oxidation reactions, or redox reactions. Oxidation is the process of matter, like an atom or ionic molecule, losing one or more electrons, and reduction is the process of the matter gaining one or more electrons. Oxidation States Each atom in a molecule has its own oxidation state or oxidation number. The oxidation state describes how oxidized a molecule is relative to its free elemental form. The oxidation state is...
