Multi-step Photolithography

Multi-step photolithography is a microfabrication method that builds precise, layered patterns on a substrate, enabling the production of complex microscale devices. The process repeats photoresist coating, mask alignment, light exposure, development, and material removal or deposition, with each exposure defining a new geometric feature and alignment preserving relationships between layers. In bioengineering, this approach supports the fabrication of microfluidic channels, biosensor structures, cell-patterning platforms, and tissue-engineering scaffolds. By controlling feature size, layout, and surface architecture, multi-step photolithography helps researchers create reproducible environments for studying cells, biomolecules, and biological transport.

This article presents a complete protocol for multi-step photolithography with valves and variable height features tunable to any application. As a demonstration, we fabricate a microfluidic droplet generator capable of producing hydrogel beads.
