T4 Dna Ligase

T4 DNA ligase is an ATP-dependent enzyme that joins DNA fragments by forming phosphodiester bonds between adjacent nucleotides, making it a foundational tool in molecular bioengineering. It first activates a DNA end with ATP and then seals a nick between a 3′ hydroxyl group and a neighboring 5′ phosphate, efficiently connecting compatible cohesive ends and, under suitable conditions, blunt ends. Researchers use T4 DNA ligase to assemble recombinant DNA, insert genes into plasmid vectors, construct libraries, and prepare engineered DNA molecules for cloning and analysis. Its ability to create stable DNA junctions supports synthetic biology, genetic engineering, and biotechnology workflows.

Amplification, Next-generation Sequencing, and Genomic DNA Mapping of Retroviral Integration Sites

We describe a protocol for amplifying retroviral integration sites from the genomic DNA of infected cells, sequencing the amplified virus-host junctions, and then mapping these sequences to a reference genome. We also describe techniques to quantify the distribution of integration sites relative to various genomic annotations using BEDTools.
