Polymer Gel Capillary

A polymer gel capillary is a narrow separation channel filled with a cross-linked or linear polymer matrix that supports electrophoretic analysis of biological molecules. When an electric field is applied, charged molecules migrate through the polymer network at different rates, largely according to size, charge, and interactions with the matrix, producing distinct separation patterns. Polymer gel capillaries are widely used in capillary electrophoresis for resolving DNA fragments, sequencing products, and other nucleic acid samples with high efficiency and small sample volumes. Their controlled format improves analytical speed, resolution, and automation in molecular biology and clinical research.

This protocol combines the characterization of a protein sample by capillary gel electrophoresis and a fast-binding screening for charged ligands by affinity capillary electrophoresis. It is recommended for proteins with a flexible structure, such as intrinsically disordered proteins, to determine any differences in binding for different conformers.

The genotyping technique described here, which couples fluorescent polymerase chain reaction (PCR) to capillary gel electrophoresis, allows for high-throughput genotyping of nuclease-mediated knockout clones. It circumvents limitations faced by other genotyping techniques and is more cost effective than sequencing methods.

In this work, we prepared an adsorbent composed of the cationic N,N-dimethylamino propylacrylamide methyl chloride quaternary (DMAPAAQ) polymer gel and iron hydroxide for adsorbing arsenic from groundwater. The gel was prepared via a novel method designed to ensure the maximum content of iron particles in its structure.
