Dna Template Synthesis

DNA template synthesis is the preparation of a defined DNA molecule that serves as a sequence-specific template for copying, transcription, or downstream analysis. It can use chemically synthesized oligonucleotides assembled into longer constructs or enzymatic amplification, in which DNA polymerase extends primers complementary to the template through repeated cycles of denaturation, annealing, and extension. In biology, synthesized templates support gene construction, PCR assay development, in vitro transcription, sequencing controls, and functional studies of regulatory or coding regions. Accurate sequence design and product verification are essential because template identity directly affects experimental reproducibility and the interpretation of molecular results.

Template Directed Synthesis of Plasmonic Gold Nanotubes with Tunable IR Absorbance

Solution-suspendable gold nanotubes with controlled dimensions can be synthesized by electrochemical deposition in porous anodic aluminum oxide (AAO) membranes using a hydrophobic polymer core. Gold nanotubes and nanotube arrays hold promise for applications in plasmonic biosensing, surface-enhanced Raman spectroscopy, photo-thermal heating, ionic and molecular transport, microfluidics, catalysis and electrochemical sensing.

Presented here is a protocol for the preparation and buffer calibration of cell extracts from exonuclease V knockout strains of Escherichia coli BL21 Rosetta2 (ΔrecBCD and ΔrecB). This is a fast, easy, and direct approach for expression in cell-free protein synthesis systems using linear DNA templates.

In this video, we determine the proliferative capacity of neuronal progenitor cells by measuring the incorporation of radioactive tracer tritiated thymidine into the DNA of cells. This technique can be used to study cell proliferation after treatment with certain drugs and growth factors.
