Model Protein Antigen

A model protein antigen is a well-characterized protein used to investigate how the immune system recognizes and responds to antigenic molecules. After uptake and processing by antigen-presenting cells, protein fragments can bind major histocompatibility complex molecules and be displayed to T-cell receptors, while intact or processed epitopes may also interact with antibodies and B-cell receptors. Researchers use model protein antigens to examine antigen processing, presentation, immune-cell activation, and antibody specificity under controlled conditions. These systems support studies of cellular immunity, vaccine design, immunological assays, and the molecular basis of host-pathogen interactions.

In this antigen-driven colitis model, OT-II CD4+ T cells expressing a red fluorescent protein were adoptively transferred into RAG-/- mice that express a green fluorescent protein in mononuclear phagocytes (MPs). The hosts were challenged with Escherichia coli (E.coli) expressing the ovalbumin protein (OVA) fused to a cyan fluorescent protein (CFP).

(2022)This video demonstrates the cell-free synthesis of nanodisc-stabilized membrane protein antigens using a dual-compartment system. The device maintains optimal conditions for efficient protein synthesis, proper folding, and stable integration into nanodiscs.

Immunostaining to Visualize Viral Antigen and Host Protein Colocalization in the Insect Gut

Source: Zhang, L., et al. Immunofluorescent Labeling of Plant Virus and Insect Vector Proteins in Hemipteran Guts. J. Vis. Exp. (2021)The video demonstrates the preparation of gut tissue from a plant virus–infected insect vector for fluorescence imaging. Fixed guts were washed, permeabilized, and incubated with two fluorescent antibodies in a blocking solution—one targeting a viral antigen in vesicles, the other a vesicle-associated membrane protein. The tissue was counterstained for actin...
