Rna Thermometers

RNA thermometers are temperature-sensitive RNA elements that regulate gene expression by converting changes in temperature into changes in RNA structure. Typically located in the 5′ untranslated region of a messenger RNA, they fold into secondary structures that can hide or expose the ribosome-binding site; temperature-dependent structural rearrangement therefore controls translation initiation. In biology, RNA thermometers help microorganisms adjust protein production during environmental transitions, including conditions associated with host infection. Their predictable regulatory behavior also supports studies of post-transcriptional control and the design of temperature-responsive systems in synthetic biology.

Thermometers and Temperature Scales

Any physical property that depends consistently and reproducibly on temperature can be used as the basis of a thermometer. For example, volume increases with temperature for most substances. This property is the basis for the common alcohol thermometer and the original mercury thermometers. Other properties used to measure temperature include electrical resistance, color, and the emission of infrared radiation. As many physical properties depend on temperature, the variety of thermometers is...

Gas Thermometers and the Kelvin Scale

The definition of temperature in terms of molecular motion suggests that there should be a lowest possible temperature, where the average kinetic energy of molecules is zero (or the minimum allowed by quantum mechanics). Experiments confirm the existence of such a temperature, called absolute zero. An absolute temperature scale is one whose zero point is absolute zero. Such scales are convenient in science because several physical quantities, such as the volume of an ideal gas, are directly...

RNA-Seq

Among different methods to evaluate gene expression, the high-throughput sequencing of RNA, or RNA-seq. is particularly attractive, as it can be performed and analyzed without relying on prior available genomic information. During RNA-seq, RNA isolated from samples of interest is used to generate a DNA library, which is then amplified and sequenced. Ultimately, RNA-seq can determine which genes are expressed, the levels of their expression, and the presence of any previously unknown transcripts.

RNA Stability

Intact DNA strands can be found in fossils, while scientists sometimes struggle to keep RNA intact under laboratory conditions. The structural variations between RNA and DNA underlie the differences in their stability and longevity. Because DNA is double-stranded, it is inherently more stable. The single-stranded structure of RNA is less stable but also more flexible and can form weak internal bonds. Additionally, most RNAs in the cell are relatively short, while DNA can be up to 250 million...
