Spore Collection

Spore collection is the controlled sampling and recovery of spores, reproductive or survival structures produced by fungi, plants, algae, and some microorganisms, for biological study. It typically involves releasing or gathering spores from a source through sterile tools, air, water, or surface-based sampling, then transferring them into labeled containers while limiting contamination and loss of viability. Researchers use collected spores to examine dispersal, germination, biodiversity, disease transmission, and environmental responses. In biology, reliable collection supports microscopy, culturing, identification, ecological surveys, and preservation of specimens for later analysis.

Source: Francis, M. B., et al. Detecting Cortex Fragments During Bacterial Spore Germination. J. Vis. Exp. (2016).This video demonstrates a colorimetric method to detect cortex degradation during bacterial spore germination by measuring the release of reducing sugars over time. Using N-acetylglucosamine standards and time-point samples, the assay quantifies sugar concentration through a purple chromogenic reaction, indicating the extent of cortex hydrolysis.
