Nuclear Lncrna Knockdown

Nuclear lncRNA knockdown is the targeted reduction of long noncoding RNA molecules that function within the cell nucleus, a strategy used to test their roles in gene regulation and cancer biology. Researchers commonly deliver complementary antisense oligonucleotides that bind a nuclear lncRNA and recruit RNase H, which cleaves the RNA-DNA hybrid and promotes transcript degradation without altering the genome. Measuring changes in transcription, chromatin-associated complexes, cell proliferation, apoptosis, invasion, or treatment response can reveal whether a lncRNA supports tumor development or suppresses it. This approach helps validate therapeutic targets and clarify regulatory mechanisms in cancer cells.

Contribution of the ACF chromatin remodeling factor to E4orf4-induced cell death was measured. The protocol includes selection of cell clones in which doxycycline treatment induces conditional knockdown of the ACF subunits Acf1 and SNF2h, and use of the DAPI assay to measure E4orf4-induced cell death in the inducible cell lines.
