Peritoneal Metastasis

Peritoneal metastasis is the spread of cancer cells to the peritoneum, the thin lining of the abdominal cavity, where secondary tumors can impair organ function and worsen prognosis. It commonly develops when tumor cells detach from a primary lesion, survive in peritoneal fluid, adhere to the mesothelial surface, and invade underlying tissue, often contributing to fluid accumulation called ascites. In cancer research, studying these steps supports the development of organoid and animal models, imaging approaches, and treatments such as cytoreductive surgery and intraperitoneal chemotherapy. This work helps clarify metastatic biology and improve disease detection, risk assessment, and therapeutic strategies.

Ovarian cancer metastasis is characterized by numerous diffuse intra-peritoneal lesions, such that accurate visual quantitation of tumor burden is challenging. Herein we describe a method for in situ and ex vivo quantitation of metastatic tumor burden using red fluorescent protein (RFP)-labeled tumor cells and optical imaging.

Modeling Ovarian Cancer Metastasis in Peritoneal Cavity Lining: A Method To Establish an In Vitro 3D Model of the Peritoneal Lining

In this video, we demonstrate the development of an in vitro three-dimensional model of the lining of the peritoneal cavity. This organotypic culture can help to investigate ovarian cancer cell adhesion, invasion, and proliferation.

To clarify the multistep process of peritoneal dissemination of ovarian carcinoma, the current study presents a murine experimental model of original tumor development and peritoneal metastasis via orthotopic inoculation with these tumor cells.

Harvesting Peritoneal Fat Depots: A Technique to Study Ovarian Cancer Colonization of Peritoneal Adipose Tissues in Mouse Models

In this video, we use a euthanized female mouse model bearing ovarian cancer. We then describe a protocol to identify and extract fat depots or adipose tissues associated with the five primary peritoneal organs to study metastatic cancer cell colonization.
