Precision-cut Tissue

Precision-cut tissue consists of thin, reproducible slices prepared from intact organs or tumors, preserving native architecture and cellular relationships for controlled ex vivo study. A specialized tissue slicer produces uniform sections under conditions that maintain tissue structure, viable cells, and components of the local microenvironment, including extracellular matrix and neighboring cell populations. In cancer research, these slices provide a biologically relevant model for examining tumor behavior, cell signaling, and responses to anticancer treatments. They can complement cell cultures and animal models by retaining features of the original tissue, supporting more representative drug evaluation and investigation of tumor–microenvironment interactions.

Organotypic primary tissue explant cultures, which include precision-cut slices, represent the three-dimensional (3-D) tissue architecture as well as the multicellular interactions of native tissue. This video describes the culturing of murine lung tumor-derived tissue slices using a rotating incubation unit, a system that enables intermittent exposure of tissues to oxygen and nutrients. The protocol describes the methodology to generate and culture precision-cut lung tumor slices for drug...

An Assay to Measure Chemically-Induced Cytotoxicity in Human Precision-Cut Lung Slices

This video illustrates a technique for evaluating cytotoxicity in human precision-cut lung slices through the WST-1 assay. Human precision-cut lung slices are subjected to incremental concentrations of a cytotoxic chemical to quantify their cytotoxicity. A decrease in color intensity serves as confirmation of the chemical's detrimental impact on lung cells.
