Spheroid Embedding

Spheroid embedding is a three-dimensional culture method that places multicellular spheroids within a supportive extracellular matrix to model tissue organization more realistically than conventional monolayer cultures. The matrix surrounds the spheroid and provides structural and biochemical cues, allowing cells to maintain cell-cell interactions, establish nutrient and oxygen gradients, and migrate or invade into the surrounding material. In cancer research, this approach helps investigators study tumor growth, invasion, cell-matrix interactions, and responses to anticancer drugs in a controlled laboratory model. Spheroid embedding can therefore improve assessment of tumor behavior and support the development of more physiologically relevant cancer therapies.

This article describes a protocol for mechanical characterisation of living spheroids within a 3D matrix using Brillouin micro-spectroscopy. This all-optical method enables spheroid visualisation with microscale resolution and quantitative mechanical properties. This approach has implications for mechanical phenotyping; for instance, determining pathological states of tumor spheroids within a 3D microenvironment.
