Basement Matrix Coating

Basement matrix coating is a laboratory method that applies extracellular matrix proteins to a culture surface to reproduce features of the basement membrane, a specialized tissue layer that supports cells and separates epithelium from underlying stroma. In cancer research, coatings typically provide adhesion sites and biochemical cues through components such as laminin and collagen, enabling tumor cells to attach, spread, and organize into physiologically relevant structures under controlled culture conditions. These models support studies of cancer-cell morphology, polarity, migration, invasion, drug response, and interactions with the extracellular matrix, while helping researchers compare tumor behavior across defined experimental environments.

Induced pluripotent stem cells (iPSCs) represent a source of patient-specific tissues for clinical applications and basic research. Here, we present a detailed protocol to reprogram human peripheral blood mononuclear cells (PBMNCs) obtained from frozen buffy coats into viral-free iPSCs using non-integrating episomal plasmids.
