Transillumination System

A transillumination system is an optical imaging setup that reveals structures by passing light through a specimen, making otherwise subtle features visible for biological and cancer research. A light source illuminates the sample from one side, while transmitted and scattered light is collected on the opposite side; differences in absorption, thickness, and optical density generate contrast. This approach can support noninvasive visualization of tissue organization, blood vessels, lesions, or developing tumor models, helping researchers document morphology and evaluate experimental changes. By providing rapid, label-free imaging, transillumination complements fluorescence and other microscopy methods when structural information is the primary outcome.

This video demonstrates the reporter enzyme fluorescence (REF) imaging for in vivo detection of Mycobacterium tuberculosis infection. The infected mice are intraperitoneally injected with a substrate for REF imaging. Upon being hydrolyzed by bacterial β-lactamase enzyme, the substrate emits fluorescence, the detection of which aids evaluation of infection.

We suggest a Born normalized approach for Optical Projection Tomography (BnOPT) that accounts for the absorption properties of imaged samples to obtain accurate and quantitative fluorescence tomographic reconstructions. We use the proposed algorithm to reconstruct the fluorescence molecular probe distribution within small animal organs.
