Hepatic Drug Metabolism

Hepatic drug metabolism is the enzymatic transformation of medicines in the liver, a process that helps determine their duration, activity, and elimination from the body. Liver enzymes, particularly cytochrome P450 proteins, commonly carry out Phase I reactions such as oxidation, reduction, or hydrolysis, while Phase II enzymes attach polar groups through conjugation to increase water solubility and support excretion. These pathways can activate prodrugs, inactivate therapeutic compounds, or produce metabolites with distinct effects. In clinical research and practice, understanding hepatic metabolism guides dose selection, predicts drug interactions, and helps explain differences in treatment response and toxicity among patients.

A progressive colonization procedure is described to further assess its impact on the host hepatic metabolism. Colonization is monitored non invasively by evaluating the urinary excretion of microbial co-metabolites by NMR-based metabolic profiling while hepatic metabolism is assessed by High Resolution Magic Angle Spinning (HR MAS) NMR profiling of intact biopsy.
