Terra Cotta Facade

A terra cotta facade is an exterior building envelope made from fired clay units that provide architectural cladding, weather protection, and visual character. Manufacturers shape and fire the clay to create durable panels, tiles, or masonry elements, which engineers attach to a supporting wall with anchors, rails, or mortar joints that accommodate movement and drainage. The system’s performance depends on material strength, water management, thermal movement, joint design, and secure connections between the cladding and structure. Terra cotta facades are used in new construction and historic restoration, offering relatively low-maintenance surfaces while supporting energy-conscious designs and preserving architectural heritage.

Here, we present a protocol to generate cancer cell clones containing a MS2 sequence tag at a single subtelomere. This approach, relying on the MS2-GFP system, enables visualization of the endogenous transcripts of telomeric repeat-containing RNA (TERRA) expressed from a single telomere in living cells.

Here, we describe a S. pneumoniae serotype 1 strain 519/43 that can be genetically modified by using its ability to naturally acquire DNA and a suicide-plasmid. As proof of principle, an isogenic mutant in the pneumolysin (ply) gene was made.
