Cellular Migration

Cellular migration is the directed movement of cells through or across tissues in response to chemical, physical, and mechanical features of their environment. Cells detect external cues such as chemical gradients, attach to the extracellular matrix through adhesion proteins, and coordinate actin cytoskeleton remodeling to extend the cell front and retract the rear. This process is essential for embryonic development, wound healing, immune responses, and tissue organization. Abnormal migration can contribute to cancer invasion and chronic inflammation, while studying how cells navigate changing environments helps researchers understand tissue function, disease progression, and strategies for regenerative medicine.

Here, we present an easy-to-use co-culture assay to analyze glioblastoma (GBM) migration on patterned neurons. We developed a macro in FiJi software for easy quantification of GBM cell migration on neurons, and observed that neurons modify GBM cell invasive capacity.

We describe a protocol for real-time videoimaging of neuronal migration in the mouse forebrain. The migration of virally-labeled or grafted neuronal precursors was recorded in acute live slices using wide-field fluorescent imaging with a relatively rapid acquisition interval to study the different phases of cell migration, including the durations of the stationary and migration phases and the speed of migration.
