Dna Extraction Method

A DNA extraction method is a laboratory procedure that isolates genetic material from cells or environmental samples for molecular analysis. It typically breaks open cells through chemical, enzymatic, or mechanical lysis, then removes proteins, cell debris, and environmental inhibitors before purifying DNA by filtration, silica binding, or precipitation. In environmental sciences, these methods recover DNA from soil, water, sediment, and other complex matrices, where substances such as humic compounds can interfere with downstream tests. The resulting DNA supports PCR, sequencing, environmental DNA surveys, microbial community profiling, biodiversity assessment, and monitoring of ecological change or pollution.

We describe here a simple and rapid paper-based DNA extraction method of HIV proviral DNA from whole blood detected by quantitative PCR. This protocol can be extended for use in detecting other genetic markers or using alternative amplification methods.

We are describing a new method of isolating genomic DNA from whole blood collected for plasma/serology. After plasma collection, the compacted blood is usually discarded. Our novel method represents a significant improvement over existing methods and makes DNA and plasma available from a single collection, without requesting additional blood.
