Alternating Excitation

Alternating excitation is a fluorescence-imaging method that switches between two or more excitation sources to distinguish signals from different fluorophores. In alternating-laser excitation, illumination cycles between wavelengths while fluorescence detection is synchronized with each pulse, allowing emission from donor and acceptor dyes to be measured separately and compared through parameters such as intensity, stoichiometry, or Förster resonance energy transfer. In genetics, this approach supports single-molecule studies of labeled DNA, RNA, and protein complexes, helping researchers characterize molecular interactions, conformational changes, binding events, and population heterogeneity with reduced spectral cross-talk.

This abstract describes a novel method to assess the development of neurotoxicity in patients receiving chemotherapy treatment. While conventional assessment methods are limited in their ability to detect early changes in nerve function, nerve excitability techniques provide early identification of patients at risk of severe neurotoxicity and insight into pathophysiology.
