CD3, CD4, and CD8 are immune-cell surface proteins that identify and distinguish T lymphocytes, making them central markers in immunology and infection research. CD3 forms part of the T-cell receptor complex and transmits activation signals after antigen recognition, while CD4 and CD8 act as co-receptors that bind major histocompatibility complex class II and class I molecules, respectively. Together, these markers classify helper T cells, cytotoxic T cells, and broader T-cell populations using flow cytometry, immunohistochemistry, or related assays. Their expression patterns help evaluate immune development, monitor infections and immunodeficiency, and characterize responses to vaccines, therapies, and inflammatory disease.

Generating Single Positive CD8 T Cells from Induced Pluripotent Stem Cells

This video demonstrates an assay to generate tumor antigen-specific induced pluripotent stem cell (iPSC)-derived CD8αβ+ T Cells. Tumor antigen-specific iPSCs are grown on a stromal cell monolayer and differentiated into hematopoietic progenitor cells (HPCs). The HPCs are differentiated into CD4+ CD8+ double-positive T cells using a differentiation medium. These cells are then stimulated to generate single-positive T cells expressing CD8.

An Assay for Cell Cycle Analysis of Antigen-Specific CD8+ T Cells

This video demonstrates an assay to analyze antigen-specific CD8+ T cells in different cell cycle stages. Fluorescently-labeled viable cells are selected using flow cytometry, and the cells in the different cell cycle stages are identified by their DNA content using antibodies against cell proliferation-associated nuclear protein and a nucleic acid stain.

We describe a standard antibody staining protocol for use in microscopy to determine the membrane expression and localization of gangliosides in resting and activated human naïve CD4+ T cells. Also described are real-time PCR experiments using <40,000 cells that do not require additional low input RNA kits.

This protocol describes the reproducible generation and phenotyping of human induced regulatory T cells (iTregs) from naïve CD4+ T cells in vitro. Different protocols for FOXP3 induction allow for the study of specific iTreg phenotypes obtained with respective protocols.
