Cell Stack Culture

Cell stack culture is a cell-culture technique that grows adherent cells across multiple vertically stacked layers within a single vessel, increasing surface area without proportionally expanding laboratory footprint. Cells attach to treated surfaces and receive culture medium within each layer, while controlled incubation supports nutrient exchange, waste removal, and sustained proliferation under consistent conditions. In immunology and infection research, this format can generate large, uniform cell populations for host-pathogen assays, immune-cell interaction studies, viral propagation, and screening experiments, helping researchers scale biological models while reducing handling and vessel-to-vessel variation.

Scaling of Engineered Vascular Grafts Using 3D Printed Guides and the Ring Stacking Method

Scalable engineered blood vessels would improve clinical applicability. Using easily sizable 3D-printed guides, rings of vascular smooth muscle were created and stacked into a tubular form, forming a vascular graft. Grafts can be sized to meet the range of human coronary artery dimensions by simply changing the 3D-printed guide size.

Here is a protocol to grow pluripotent stem cells (PSC) and neural stem cells (NSC) in an enclosed cell culture system that permits maximum sterility and reproducibility, replacing the traditional biosafety cabinet and incubator. Here we provide a detailed procedure for large-scale production of research-grade AAV vectors using adherent HEK 293 cells grown in cell stacks and affinity chromatography purification. This protocol consistently yields >1 x 1013 vector genomes/mL, providing vector quantities appropriate for large animal studies.
