Percoll Gradient

A Percoll gradient is a density-based separation method that fractionates cells, organelles, or microorganisms according to their buoyant density, making it valuable for preparing purified samples for research. It uses centrifugation through layers of Percoll, a colloidal silica suspension coated with polyvinylpyrrolidone, so particles migrate until they reach positions matching their density. In immunology and infection research, Percoll gradients can enrich leukocyte populations, separate immune cell subsets, and help recover pathogens or infected cells from complex biological samples. The resulting fractions support downstream flow cytometry, microscopy, culture, and functional assays by reducing cellular debris and sample heterogeneity.
