Basic Calcium Phosphate

Basic calcium phosphate (BCP) is a family of poorly crystalline calcium phosphate minerals, including hydroxyapatite, that normally contribute to hard-tissue structure but can also accumulate abnormally in soft tissues and joints. These crystals form when local calcium and phosphate concentrations, tissue conditions, or matrix properties favor mineral deposition; once deposited, they may be taken up by immune and resident cells, activating inflammatory signaling and tissue-damaging responses. In medicine, BCP is relevant to calcific tendinopathy, periarthritis, cartilage degeneration, and some destructive arthropathies, where identifying crystal deposits helps clarify disease mechanisms and informs imaging, laboratory investigation, and treatment strategies.

In this video, we describe an in vitro osteoclastic resorption assay on calcium phosphate-coated cell culture plates cultured with human peripheral blood mononuclear cell-derived osteoclasts. The osteoclasts form a characteristic actin ring on the calcium phosphate-coated matrix of the cell culture plate and degrade the matrix within the actin ring region to release calcium and phosphate ions, resulting in the formation of distinct resorption pits within the degraded matrix.
