Intratumoral Injection

Intratumoral injection is a medical technique that delivers a therapeutic substance directly into a solid tumor, concentrating treatment at the disease site while potentially limiting exposure to healthy tissues. Using a needle placed within the tumor, clinicians deposit drugs, biological agents, or investigational therapies into tumor tissue, where local diffusion and agent-specific effects can damage cancer cells, alter the tumor microenvironment, or stimulate an immune response. The procedure may be guided by imaging and is used in selected treatments and clinical research, including studies of immunotherapies and combination regimens. By enabling localized treatment and direct assessment of tumor response, intratumoral injection supports precision oncology and therapy development.

Modelling Intratumoral Treatment with miRNA Inhibitors: A Method to Analyze the Effect of miRNA Inhibitors on Tumor Growth in Murine Model

2023

This video demonstrates the in vivo delivery of miRNA inhibitors as therapeutic agents against tumor growth in orthotopic mouse models. The intratumoral administration of synthetic miRNA inhibitors or antagomiRs allows them to bind and inhibit endogenous miRNAs, thereby causing mRNA upregulation and suppression of tumor growth.

2025

This protocol presents techniques and methodology necessary for targeted hyperthermic therapy in solid tumor models. The approach leverages the photothermal conversion of near-infrared light by intratumorally injected gold nanorods to induce localized heating within the tumor microenvironment.

Endobronchial Ultrasound-guided Intratumoral Injection of Cisplatin for the Treatment of Isolated Mediastinal Recurrence of Lung Cancer

The management of isolated recurrent lung cancer in a previously-irradiated field is challenging. Here, we describe an endobronchial ultrasound (EBUS)-guided cisplatin injection for the management of patients with localized lung cancer recurrence in a previously-radiated field.

Intradermal Tumor Implantation as a Robust Model for Murine Cancer Intratumoral Immunotherapy Studies

2026

With increasing interest in intratumoral immunotherapy, selecting tumor models that are accessible and practical is essential. Subcutaneous tumors are easy to establish but difficult to visualize and manipulate. In this protocol, we describe methods for generating intradermal tumors that provide improved visibility and accessibility.

After neural tube formation, the neuroepithelium constricts and folds while the tube fills with embryonic cerebrospinal fluid (eCSF) to form the embryonic brain ventricles. We developed this ventricle injection technique to better visualize the fluid filled space in contrast to the neuroepithelial shape in a live embryo.
