Macrophage Infiltration Assessment

Macrophage infiltration assessment is the measurement of macrophage presence, density, and distribution within tissues, helping characterize immune activity in disease. Researchers typically identify macrophages in tissue sections using immunohistochemistry or immunofluorescence markers such as CD68 or CD163, then quantify labeled cells or staining intensity across defined regions. In medicine, this approach supports evaluation of inflammation, infection, tissue injury, and tumor microenvironments, where macrophage abundance and localization can relate to disease progression or treatment response. Standardized image analysis and complementary molecular methods improve reproducibility, enabling comparisons among patient samples and strengthening studies of immune-mediated pathology and therapeutic targets.

Immunohistochemistry to Study the Relationship Between Macrophages and Infiltrated Immune Cells

2025

This video demonstrates a technique to quantify the relative ratio of macrophages among the total infiltrated immune cell populations in temporal artery biopsy. The immunohistochemical staining of a tissue section affected by autoimmune disease selectively stains the immune cells with specific receptors to visually differentiate them from the rest of the cells.

Assessment of Leishmania Virulence within Cultured Macrophages

2025

This video investigates Leishmania's virulence by treating macrophages with metacyclic promastigotes, resulting in the formation of intracellular amastigotes. The confirmation of Leishmania's virulence is achieved through nucleic acid fluorescent dye staining. This insight can contribute to the development of new therapies for leishmaniasis.

Investigation of Macrophage Polarization Using Bone Marrow Derived Macrophages

The article describes a readily easy adaptive in vitro model to investigate macrophage polarization. In the presence of GM-CSF/M-CSF, hematopoietic stem/progenitor cells from the bone marrow are directed into monocytic differentiation, followed by M1 or M2 stimulation. The activation status can be tracked by changes in cell surface antigens, gene expression and cell signaling pathways.

Assessment of the Activation of Multiple Caspases in Macrophages

2025

This video demonstrates the assessment of multiple caspase activations in bone marrow-derived macrophages through stimulation with the influenza virus. Following macrophage lysis, heat-inactivation of lysate, and centrifugation, intracellular caspases are collected for subsequent immunological analysis.

Source: Zhang, C. et al. Development and Assessment of Intracellular Infection Models for Staphylococcus aureus. J. Vis. Exp. (2025)This video demonstrates the quantification of intracellular survival of a methicillin-resistant bacterial pathogen in mouse peritoneal macrophages. It outlines the steps involved in infection, extracellular bacterial clearance, host cell lysis, and colony enumeration to assess bacterial survival within host macrophages.
