Roi Signal Subtraction

ROI signal subtraction is an image-analysis method that removes background or reference signal from a defined region of interest (ROI) to isolate the measurement relevant to a medical investigation. The process typically involves selecting the ROI, measuring its observed intensity, estimating signal from a background or control region, and subtracting the reference value to reduce nonspecific contributions. This approach can improve contrast and support more reliable quantitative interpretation of medical images, including comparisons among tissues, lesions, or treatment conditions. By reducing background variation, ROI signal subtraction helps researchers assess subtle signal changes and strengthen reproducibility in imaging-based studies.

Rapid Subtractive Patterning of Live Cell Layers with a Microfluidic Probe

We present a protocol to perform subtractive patterning of live cell monolayers on a surface. This is achieved by local and selective lysis of adherent cells using a microfluidic probe (MFP). Here a novel region of interest analysis protocol based on sorting best-fit ellipses assigned to regions of positive signal within two-dimensional time lapse image sequences is demonstrated.
