Split-mouth Trial

A split-mouth trial is a controlled clinical study in which different treatments are assigned to separate, comparable sites within the same participant, allowing each person to serve as their own control. Researchers typically randomize the side or site receiving each intervention, then compare outcomes such as healing, pain, disease progression, or treatment response under similar patient-level conditions. This paired design can reduce variability from age, genetics, and overall health, improving statistical efficiency with fewer participants. It is especially useful when conditions affect multiple symmetrical sites, although researchers must account for treatment carryover, interactions between sites, and the risk that one site’s treatment influences another.
