Tissue Marker Delivery

Tissue marker delivery is the process of transporting detectable labels to specific tissues so their structure, cellular features, or disease-associated targets can be identified. Depending on the marker and delivery strategy, the label may reach tissue through local administration, systemic circulation, or a carrier that promotes selective uptake, then bind to a molecular target or accumulate in a defined region for detection. In medicine, tissue marker delivery supports diagnostic imaging, histopathology, biomarker validation, and treatment-response assessment. Improved targeting and controlled distribution can increase signal quality, reduce off-target labeling, and strengthen the ability to distinguish healthy from abnormal tissue.

The video demonstrates a method for directly transferring the interleukin-15 encoding plasmid DNA into the skin tissue of an interleukin-15 deficient mouse model through acupuncture using microneedles. These microneedles generate micropunctures in the outermost skin layer, facilitating the absorption and transfection of plasmid DNA into the epidermal cells, including keratinocytes, within the skin tissue.

For creation of highly organized structures of complex tissue, one must assemble multiple material and cell types into an integrated composite. This combinatorial design incorporates organ-specific layered cell sheets with two distinct biologically-derived materials containing a strong fibrous matrix base, and endothelial cells for enhancing new vessels formation.
