Cell Aggregate Dissociation

Cell aggregate dissociation is the process of separating multicellular clusters into individual cells or smaller populations while preserving cell viability and biological function. It works by combining mechanical disruption with chemical or enzymatic treatment that weakens cell-cell adhesion and extracellular matrix components, with exposure time and handling conditions adjusted to limit cellular damage. In neuroscience, dissociation of neural spheroids, organoids, or tissue aggregates enables single-cell culture, cell counting, flow cytometry, and molecular profiling. The resulting suspensions support studies of neuronal development, glial biology, disease mechanisms, and treatment responses, although excessive processing can alter cell morphology, viability, and gene expression.

4D Imaging of Protein Aggregation in Live Cells

Cellular viability depends on timely and efficient management of protein misfolding. Here we describe a method for visualizing the different potential fates of a misfolded protein: refolding, degradation, or sequestration in inclusions. We demonstrate the use of a folding sensor, Ubc9ts, for monitoring proteostasis and aggregation quality control in live cells using 4D microscopy.

Tracking Hypoxic Signaling within Encapsulated Cell Aggregates

A method for photo-encapsulation of cells in a crosslinked PEG hydrogel is described. Hypoxic signaling within encapsulated murine insulinoma (MIN6) aggregates is tracked using a fluorescent marker system. This system allows serial examination of cells within a hydrogel scaffold and correlation of hypoxic signaling with changes in cell phenotype.
