Double Injection Technique

The double injection technique is a neuroscience method that uses two targeted injections to examine neural connections, pathways, or responses across brain regions. Researchers place distinct tracers, dyes, drugs, or other experimental agents into selected sites, then compare their distribution or effects to determine anatomical connectivity and functional relationships. By controlling injection location, volume, and timing, the method can reveal whether neuronal populations share projections, respond to related signals, or contribute differently to a circuit. Double injections support neuroanatomical tracing, circuit mapping, and studies of brain organization, providing a foundation for investigating connectivity in health, disease, and experimental models.

Direct injection into the rat optic nerve is useful for regenerative research. We demonstrate a minimally-invasive technique for direct injection into a rat optic nerve that does not involve opening the skull. Using this method, surgical complications are minimized and recovery is more rapid.

We described in this protocol a standardized subarachnoid hemorrhage (SAH) mouse model by a double injection of autologous whole-blood into the cisterna magna. Facilitating Repeat Intracarotid Injections in Mouse Models by a Novel Injection Site Repair Technique

Repair of the intracarotid artery in a mouse model after injection returns blood flow to the artery without negatively impacting the distribution of the injected material. Injection site repair facilitates subsequent injections through the same artery and prevents cerebral ischemia in mouse strains that lack a complete Circle of Willis.
