Drosophila Traumatic Brain Injury

Drosophila traumatic brain injury (TBI) is a laboratory model of mechanically induced neural damage that helps researchers study how the nervous system responds to injury. In this model, controlled impacts to the fly’s head produce acute neuronal dysfunction and can trigger blood-brain barrier disruption, neuroinflammation, oxidative stress, and progressive behavioral impairment. Because Drosophila offers well-characterized genetics, a compact nervous system, and rapid reproduction, researchers can investigate injury mechanisms and test candidate genes or interventions efficiently. Findings from this neuroscience model can clarify conserved pathways involved in TBI and support development of approaches for studying neural repair and long-term neurological consequences.

The video demonstrates the use of a High-Impact Trauma (HIT) device to induce closed-head traumatic brain injury (TBI) in Drosophila. TBI causes mechanical brain trauma affecting neuronal function, which over time leads to neurodegeneration and mortality.

Here we describe a method to inflict closed head traumatic brain injury (TBI) in Drosophila. This method provides a gateway to investigate the cellular and molecular mechanisms that underlie TBI pathologies using the vast array of experimental tools and techniques available for flies.

Inducing Penetrating Traumatic Brain Injury in Adult Drosophila Flies

This video demonstrates the induction of penetrating traumatic brain injury in the drosophila brain. Newly emerged flies are selected, anesthetized, and positioned under a fluorescence-equipped stereo microscope. The GFP-expressing mushroom body is identified, and a Minutien pin is inserted to cause injury. The flies are allowed to recover for further studies.

Athletes absorb several hundred mild traumatic brain injuries (mTBI)/concussions every year; however, the consequence of these on the brain is poorly understood. Therefore, an animal model of single and repetitive mTBI that consistently replicates clinically relevant symptoms provides the means to advance the study of mTBI and concussion.
