Egfp Luciferase Expression

EGFP luciferase expression refers to producing enhanced green fluorescent protein and luciferase as reporter molecules to reveal when and where a gene is active. Regulatory DNA drives transcription and translation of the reporter genes; EGFP emits green fluorescence after excitation, while luciferase catalyzes a substrate-dependent bioluminescent reaction, providing complementary optical readouts. In neuroscience, these reporters help visualize labeled neurons, monitor transgene or promoter activity, and track molecular changes across cells, tissues, or living preparations. Combining fluorescence with bioluminescence can link cellular localization to quantitative measurements of gene expression, supporting studies of neural development, circuit function, and disease-related regulation.

2012

Circadian clocks function within individual cells, i.e., they are cell-autonomous. Here, we describe methods for generating cell-autonomous clock models using non-invasive, luciferase-based real-time bioluminescence technology. Reporter cells provide tractable, functional model systems for studying circadian biology.

Here, we describe the methodology used for generating L. panamensis and L. donovani strains expressing the gene for eGFP as a stable integrated transgene using the pLEXSY system. Transfected parasites were cloned by limiting dilution, and clones with the highest fluorescence intensity in both species were selected for further use in drug screening assays.
