Image-guided Microscopy

Image-guided microscopy is a technique that combines microscopy with visual information to locate, target, and monitor specific structures during image acquisition. It uses recorded images, spatial coordinates, and real-time feedback to guide microscope positioning, adjust the field of view, or direct measurements toward selected biological features. In neuroscience, this approach supports precise imaging of neurons, neural circuits, and cellular activity across complex tissue, helping researchers track the same structures over time and relate microscopic changes to anatomy or function. By improving targeting and reproducibility, image-guided microscopy strengthens quantitative studies of brain organization, development, and disease.

The bottleneck for cellular 3D electron microscopy is feature extraction (segmentation) in highly complex 3D density maps. We have developed a set of criteria, which provides guidance regarding which segmentation approach (manual, semi-automated, or automated) is best suited for different data types, thus providing a starting point for effective segmentation.

Source: Peiman Shahbeigi-Roodposhti and Sina Shahbazmohamadi, Biomedical Engineering Department, University of Connecticut, Storrs, Connecticut Optical microscopes have been around for centuries, and while they reached their theoretical limitation of resolution decades ago, new equipment and techniques, such as confocal and digital image processing, have created new niches within the field of optical imaging. The best optical microscopes will typically have a resolution down to 200 nm in ideal...
