Multi-unit Activity

Multi-unit activity is the aggregate electrical signal generated by action potentials from multiple nearby neurons, providing a population-level view of neural activity in neuroscience. During extracellular recording, an electrode detects voltage changes in surrounding tissue; signal-processing methods can separate waveforms into individual units or quantify the combined multi-unit signal across time, behavioral events, or experimental conditions. Researchers use these measurements to examine coordinated firing, sensory responses, network dynamics, and changes in neural populations when single-neuron resolution is difficult. Multi-unit activity therefore links cellular electrophysiology with circuit-level function in both basic and applied research.

This protocol describes a method to obtain in vivo, high-density single-neuron recordings from the brainstem of head-fixed mice. This approach is deployed to measure the action potential firing of neurons in the ventrolateral periaqueductal gray - a brainstem region inactive during Rapid Eye Movement (REM) sleep - before and during general anesthesia.
