Neuronal Tracer Injection

Neuronal tracer injection is a neuroscience technique that labels neurons and their connections, allowing researchers to map the organization of neural circuits. A tracer is introduced into a selected brain region or neuron, then transported along axons by cellular transport mechanisms; anterograde tracers reveal projections away from the injection site, whereas retrograde tracers identify neurons that project toward it. After tissue processing, labeled cells and fibers are visualized using microscopy to determine pathway structure and connectivity. This approach supports studies of brain anatomy, sensory and motor pathways, disease-related circuit changes, and the functional organization of the nervous system.

The efficacy of intramuscular uptake and retrograde transport of molecules to corresponding motor neurons depends on the location of the injection sites with respect to the motor end plates (MEPs). Here, we describe how to locate MEPs on skeletal muscles to optimise retrograde transport of tracers into motor neurons.

Electrophysiological characterization of neuronal responses is important for understanding brain function and for guiding the placement of dyes for pathway tracing. However, many studies are performed in anesthetized animals. To understand brain function without anesthetics, we developed a method to record neuronal response properties and inject dyes in awake mouse.

An easy and convenient method to determine the extent of gap junction tracer coupling between retinal neurons is described.
