Postnatal Mouse Brain

The postnatal mouse brain is the developing brain of a mouse after birth, providing a tractable model for studying how neural circuits mature and adapt. During this period, neurogenesis, synaptic formation and pruning, myelination, and changes in gene expression progressively shape brain structure and function in response to intrinsic programs and sensory experience. Neuroscience researchers use postnatal mice to examine developmental processes, cell signaling, circuit connectivity, behavior, and responses to injury or disease. Findings from these studies help clarify mechanisms of brain maturation and support research into neurodevelopmental disorders, learning, plasticity, and potential therapeutic strategies.

In vivo Postnatal Electroporation and Time-lapse Imaging of Neuroblast Migration in Mouse Acute Brain Slices

Neuroblast migration is a fundamental event in postnatal neurogenesis. We describe a protocol for efficient labeling of neuroblasts by in vivo postnatal electroporation and subsequent visualization of their migration using time-lapse imaging of acute brain slices. We include a description for the quantitative analysis of neuroblast dynamics by video tracking.

Mosaic Analysis of Gene Function in Postnatal Mouse Brain Development by Using Virus-based Cre Recombination

An in vivo method to test gene function in postnatal brain is described. Recombinant AAVs expressing Cre and/or a fluorescent protein are injected into neonatal mouse brain. Mosaic gene inactivation and sparse neuronal labeling are achieved, allowing rapid analysis of gene function in processes critical to neural circuit development.
